Aegilops tauschii Cosson is the donor species of D genome in common wheat,it is widely distributed in the world.It contains many genes that has excellent resistance to diseases, insect pests and cold tolerance and has many other excellent qualities..Thus, it is a valuable genetic resources to ameliorate In the middle reaches of the Yellow River , there are many kinds of Aegilops tauschii, Some of them have been studied, but without a large-scale investigation. The purpose of this study is to investigate the combinations of the HMW-GS in Aegilops tauschii in the middle reaches of the Yellow River, and to find the valuable germ plasm in Ae. Tauschii.In recent years studies have shown that, the transference of plant disease-resistant genes from the relatives is an important way to expend disease-resistant genetic basis of wheat varieties. Studies on powdery mildew, rust, scab resistance has made at home and abroad in wheat plants. However, the identification of sheath blight resistance is still rarely reported. Therefore, identification of the wheat plant's resistance to sheath blight from relatives from relatives is of great significance.500 accessions of Ae. tauschii from the middle reaches of the Yellow River were analysed by SDS-PAGE A-PAGE and AS-PCR technology on this study.The purporse of this study is estimate the diversity of this Aegilops tauschii. It can offer base for distent hybrid. Identification of resistence to Wheat Sharp eyespot were carried on in this Ae. tauschii. The main result is as follows:1.Composition analysis of HMW-GS:Acollection of 500 accessions of Ae. tauschii was investigated for the allelic composition of the high molecular weight subunits at the Glu-Dt 1 locus by SDS-PAGE analysis.Threex-type Glutenin subunits (5~t,6.2~t,null) and Threey-type subunits (10.5~t,10.4~t,10.3~t), were identified,respectively,according to their ascending mobility .Among them, subunit 6.2~t was a novel x-type glutenin subunit.Three HMW-GS combinations (5~t+10.5~t ,6.2~t +10.4~t, null+10.3~t,)were found in these collection,of which their percent were 96.95%,2.44% and 0.16%. The results indicated that some genetic diversity were presented in the 500 accessions of Aegilops tauschii.2.AS-PCR: Tiriticum aestivum L. and Ae. tauschii which contain the dx-5 subunit were amplified using the dx-5 marker.The results shown that there is no distinguish between them in 1.5% agar gel.3.A-PAGE:there is one type in this collections of Ae. Tauschii.4.Evaluation of Resistance to Sharp Eyespot:The immunie ratio is 35.9%.High resistence ratio is 25.8%.So they have a fine resistence to wheat Sharp Eyespot.
