Potato is a world-cultured crop and ranked 4th in the production following wheat, maizeand rice. It also is an important crop as food, feed, processing and inductrial material, andwidely cultured in China. However, in potato culture, diseases often cause huge loses.Creating a new broad-spectrum resistant variety has become the goal of potato breeders.The hypersensitive response-assisting protein (HRAP) gene is one of the important HCD(hypersensitive cell death) associated genes that could be used for protecting plants frombacterial pathogen attack. The introduction of hrap gene into potato cultivars couldimprove the resistance of the cultivars to diseases. In this study, the hrap gene wasintroduced into poatao by means of Agrobacterium-mediated transformation.The transformation of plants was carried out using Agrobacterium tumefaciensLBA4404-based vactor containing a hrap gene. Agrobacterium was cocultured with theleaf discs from seven different varieties: Zhongshu No1, Zhongshu No3, Dongnong 303,Atlantic, Favorita, Russet Burbank (America) and Russet Burbank (Canada). The callusand shoots were induced and a total of 40 regenerated lines including 261 plants wereobtained. The PCR analysis was performed with these plants. PCR and showed that thereare 163 positive plants belonging to 36 transgenic lines. Southern blot analysis confirmedthat the hrap gene has been integrated into the genome of 12 transgenic lines. The diseaseresistance assays showed that 7 plants had resistance to the poatao late blight, and 10individual plants had resistance to bacterial wilt.
