Many clinical practices show that the prescription of Chinese medicines which contain Herba Epimdiiis are good for the intention of bone fracture and the treatment of bone metabolic diseases. The poupose of this experiment was to investigate the pharmacological founction of Icariin which was the primary component of Herba Epimdii on the chicken embryo osteoblast and Osteoclast form in vitro so that to explore the effect of icariin on the chicken bone metabolism and offer theoretic supports to put Herba Epimdii into treating and proventing Cage Layer Osteoporosis.1 Inducing culture of Osteoclast-like cell from chicken embryo marrowThe aim of this experiment was to find a simple and effective inducing culture system of the osteoclast-like cells from the embryonic chicken in order to supply a new way to study the effect of icariin on chicken osteoclast and establish a standard experimental level for the cage layer osteoporosis. Tibias and humeri were excised from the chicken embryos(18 days of age) and cleaned of extraneous soft tissue. The bone marrow was washed withα-MEM using injector. The cell supematants obtained were inoculated on culture dishs. The nonadherent cell was collected after two hour and resuspend to make the cells be 2×106/mL. Then cell supematants inoculated again, during which the 1,25-Dihydroxyvitamin D3 and dexamethasone were added wells at different concentration. Osteoclasts-like cells were successfully obtained with features of multinucleated giant size, positive staining for TRAP and bone absorptive lacunae on the bone slice.2 Effects of icariin on Proliferation and Differentiation of Osteoblasts from Chicken EmbryoThe osteoblasts isolated from the embryonic chicken was cultured to study the effects of Icariin on proliferation and differentiation of the osteoblasts. Different dosages of Icariin were added into osteoblasts cultured in vitro, the effects of Icariin on the osteoblasts were observed at 24 h, 48 h and 72 h. The results showed that Icariin could promote the osteoblasts differentiation and slightly promote proliferation. The effects of Icariin were relative to the dosage of Icariin. As the Icariin concentration was 20 ng/mL, the effect was obvious3 The effect of Icariin on the formation and function of the Osteoclast-like cells Based on the methods of inducing culture system of the osteoclast-like cells from the embryonic chicken, the concentration of Icariin were choosed based on the experimentⅡresultsto observe the effects of Icariin on the formation and function of the osteoclasts. Different concentrations of Icariin were added to observe the effects of Icariin on the formation of the osteoclasts-like cell, the results showed that Icariin could significantly depress the number of osteoclasts-like cells induced by 1, 25-Dihydroxyvitamin D3 and dexamethasone. At the eighth day, as the osteoclasts-like cell formed, different concentrations of Icariin was added to observe the effects of Icariin on the function of osteoclasts-like cells. The results showed that.
