Canine distemper(CD)caused by canine distemper virus (CDV) is an acute infectious disease which is characterized by high pathogenicity and infectivity. It is imminent to prevent CD for its high mortality to canine, fur animal and wildlife, etc.Monoclonal antibody against canine distemper virus nucleoprotein was developed by means of the conventional protocol.After immunization of BALB/c mice with purified canine distemper virus (CDV),six hybridoma cell strains against CDV were developed after fusion between SP2/0 myeloma cell and the stimulated splenocytes. The indirect ELISA results showed that the McAbs ascites titer were 1×10~7,1×10~7,1×10~7,1×10~5,1×10~4,1×10~4.No cross reaction was found when McAbs reacted with RV, CPV and CAV.The result of western-blotting with purified CDV is positive, which indicates these monoclonal antibodies all aim directly at canine distemper virus nucleoprotein.The six McAbs are were purified by protein G and labelled with FITC. The results of absorption test and blocking test showed that the fluorescent antibody (FA) has highly specificity and sensitivity , and has no interaction with CPV, RV and CAV. The IgG was purified by affinity chromatography and conjugated with FITC ,CDV in cell were tested by using direct immunofluorescent assay. The results show this established method is of rapid and high specific for diagnosis of CDV.
