The Eucommia ulmoides is the Eucommiaceae plant,it is our country precious nutritious raw material for medicine,it has many effects such as tonifying liver and kidney, strengthen muscles and bones,falls the blood pressure,soothing the fetus and so on.Its for medicinal purposes history is glorious,has the widespread application in clinical.The polyploid plant possessed gigantism,the induction cultivation Eucommia ulmoides polyploid plant can increase Eucommia ulmoides leaf,the dark output,having the important value to Eucommia ulmoides bumper crop cultivation.This research mainly uses the colchicin processing Eucommia ulmoides seed growing point,has studied the colchicin solution density,the process time mutafacient effect,through the stem point cell chromosome number examination and the DNA comparative content determination ploidy appraisal method,obtains the Eucommia ulmoides tetraploid plant for the first time;Buds in the seed in the colchicin processing Eucommia ulmoides,explored the processing effect of the different radicle length seed has been different,and to processed after under three kind of different raise conditions the seed morphological change rate and the survival rate has carried on the synthesis comparison.The main research results were as follows:1.In the colchicine processing Eucommia ulmoides seeding,rearch the optimum treatment combination with different concentrations of colchicine solution at different stages.It was 0.1% colchicine solution for 12 h,and the rates of vatiant plants and tetraploid plants were 63.3%and 36.7%,respectively.2.Basing on the morphological and anatomic study,The plants showed the general characteristics of tetraploids,such as,thicker leaf,smaller leaf index,larger guard cells, less stomata per unit area and more chloroplasts in guard cells;The width of the palisade tissue and the sponge tissue increase,The cells of the stem point meristematic tissue level correspondingly are larger.3.According to the cytological study,the chromosome number in the polyploid plants are 2n=2x=34,but in the mutants are 2n=4x=68;Determination the DNA comparative content of the cell by cytometer the result showed that the DNA content in the cells of the polyploid plant increased one time,turned 4c by 2c and it has not seen the octoploid cell.4.Eucommia ulmoides germinated seeds are mutagenized by colchicin.The radicle length is longer to its growth more disadvantageous,it should choose the slightly short radicle and the suitable time.In the same time(12h),the mutant effect of the 0.5cm radicle length is good in the 0.05%～0.1%colchicine density.5.Comparison the three kind of raises condition(tissue culture,incubator raise,plant nursery raise),the tissue culture and the incubator raise induction effect is close,and surpasses the plant nursery raise;Processing 12h by 0.5%～0.1%colchicine density the effect to be best.
