This research was study on not only birth, 30, 60, 70, 90days in female Xiao Meishan pig and birth, 60, 120, 150, 180 days in female Sujiang pig. We used immunohistochemical medthod and fluorescent quantitation PCR (FQ-PCR) technology, aimed to associate the disposition and localization in hypothalamus, pituitary and ovary of KISS-1, which was related to puberty, with the change of development expression, probed the function of this gene in puberty initiation mechanism, provided rationale for illuminating animals reproduction mechanism and selection of prematurity breeds. The results were followed:1. The disposition and localization of KISS-1mRNA in hypothalamus, pituitary and ovary were detected by immunohistochemical medthod. KISS -1 mRNA positive particles widely distributes in hypothalamus, the ovary, in the pituitary , the nucleus's positive particles are most especially obvious in the 70 days; In all levels of graafian follicles are most by mature follicle's positive particles.2. At five developmental stages, the concentration rules of FSH, LH, E2, P4 in blood plasma were detected by radioactive immunoassay. The results showed that: the concentration rules of FSH, LH, E2, P4 in blood plasma in Sujiang were almost coincided with Meishan. Showed high level at birth, then descended quickly, began to ascend before puberty (60 days in Meishan sow, 120 days in Sujiang sow), achieved the peak value at puberty. But the concentration of P4 was low, and was steady before its crest-time, the difference was not significant(P>0.05).3. The change expression abundance of KISS-1 mRNA in hypothalamus, pituitary and ovary detected with FQ-PCR technology in two species sows at five different developmental stages. The results suggested that: it was maximal at puberty of three tissues, ascended from birth to puberty and descended after puberty. But compared with Xiao Meishan sow, the change expression abundance of KISS-1 mRNA in Sujiang was slow-moving,but it's difference form every apparatus.At different developmental stages, in Xiao Meishan sow, the difference between hypothalamus and ovary was not significant (P>0.05), it was significant(P<0.05)between hypothalamus and pituitary, and it was significant (P<0.05)between pituitary and ovary. In Sujiang sow, the differences between three tissues were all not significant (P>0.05). But it was significant(P<0.05)between pituitary and ovary.The research approached the relationship between KISS-1 gene and puberty initiation in pig, determined localization of KISS-1 mRNA in HPG, the expression rules of KISS-1 mRNA before and after puberty, and the change rule of hormone in Sujiang and Xiao Meishan. The results showed that: the hybridization signals KISS-1 mRNA in HPG and the expression abundance were coincided with the change of hormone before and after puberty, which confirmed that KISS-1 gene was the key gene for initiation of puberty.
