DNA Polymorphism Of The Genes Relating To Lipid Biosynthesis Between Tapidor And Ningyou-7, The Two Parents For The T-N Mapping Population

Investigated are the DNA polymorphism of the lipid genes between Tapidor(T) and Ningyou-7(N),the two typical cultivars for European and Chinese winter rapeseed(Brassica napus),respectively.The rapeseed EST sequences were obtained by the search of public database on the baisi of the known sequences of the Arabidopsis orthologous genes by BLAST.The main results are described as the following.1.57 EST sequences of the genes that are preferably expressed in seed were found based on the knowledge of the sequences of the arabidopsis orthologous genes and the search in the NCBI database;2.150 pairs of primer were designed according to the EST sequences of rapeseed lipid genes.71 molecular markers have been identified,which revealed the DNA polymorphisms between Tapidor and Ningyou-7.76.5%of the markers were found in the genes that were preperably expressed in seed.3.Three types of the molecular markers have been roughly classified.These are length polymorphism,dominant/recessive polymorphism,and the polymorphism revealed by the both.The statistic data showed that more polymorphisms could be recognized in the 5' end region of the genes,and more dominant/recessive type than length polymorphism type markers could were found;4.Several experimental methods,as like SDS-PAGE,SSCP,HAD-GT12^TMmulti capillary electrophoresis system,Megabase-1000 genotyping capillary system, were compared for the efficiency in finding DNA polymorphisms,given cost factor.The markers found can be further applied in analyzing the genotype of the 202 T-N DH lines,thus localizing the molecular markers on the existing T-N map.
