Compounds Fructus Mume is a kind of fishery drug to prevention and cure aquatic Livestock's bacteroidal disease.There are much of efficient components such as Citric Acid, malic acid, gallic acid and baicalin,which have the effect to antibiotic, detoxificate and restorate.They have been used in aquatic products to prevention and cure bacteroidal disease.However ,there have been no studies about the systerm the pharmacokinetics and the pharmacodynamics of Compounds Fructus Mume in aquatic Livestock.The vitro antibiosis and bacteriostasis effections were verified in the dissertation; In the dissertation, the methods of Ultrasonic Extraction and assay methods of contents with HPLC Were established; The effections of Liver function and anti-aging were reflection by physiological and biochemical indicator in johnny carp's blood with giving bacterium and Compounds Fructus Mume; The pharmacokinetics and bioavailability were studied in johnny carp, reference datas for route of administration were suplied with the experiments.1. Studies on pharmacokinetics of Compounds Fructus Mume1.1 Studies on the Ultrasonic Extraction of effective constituentTo investigate the optimal preparation technique of ultrasound on the extraction of Compounds Fructus Mume, Orthogonal method was used in this shudy with organic acid and total flavonoids content for indices. The suitable preparation technique: powder with water(1:20)for 15min(×3) in the temperature of 60℃.The organic acid and total flavonoids in prescription Fructus Mume contents were 19.14% and 27.56‰. 1.2 Content determination of effective components in Compound Frunus mume. by HPLCObjective: To establish the simultaneous determination method of Citric Acid, malic acid, gallic acid and baicalin. Method: Using HPLC method as the determination method. The mobile phase was Acetonitrile - H3PO4water solution (pH 2.55) . Adopted linear gradient elution, At the time of 0~20 min, 5%~15%,and detected at 215nm with flow rate of 0.6mL;At the time of 20~60min, 15%~45%,and detected at 280nm with flow rate of 0.6mL.Result:The content of Citric Acid, malic acid, gallic acid and baicalin were 86.39‰,31.93‰,6.60‰and 7.61‰.Conclusion:The method was convenient stable reliable and special for the dermination of Compounds Frunus mume.1.3 The pharamacokinetics study of Compounds Frunus mume in johnny carpThe drugs were given through mouth and intramuscular injection. Choosing the gallic acid as the target,the changes of the gallic were tested by HPLC in the johnny carp'blood.The metabolism regularities and bioavailability were reflect with the pharamacokinetics studies ,they were conduce to Medication program.The datas were dealed with 3P97 pharamacokinetics software,and the regularities of absorption, disposition and annihilation were reflected. In the two group, the curves between plasma concentration of drug and time could be described by pen one-compartment model ,the equation with mouth was C=21.8244e-0.1732t and with intramuscular injection was C=15.1413e-0.0744t .In Compounds Frunus mume withmouth,AUC(0-∞)=105.915μg/(mL·h),Cmax=12.956μg/mL,t1/2Ka=0.637h,t1/2Ke=4.001h..In Compounds Frunus mume with intramuscular injection,AUC(0-∞)=203.395μg/(mL·h),Cmax=22.200μg/mL,t1/2Ke=9.311h, and absolute bioavailability F=52.074%.2. Studies on pharmacodynamics of Compounds Fructus Mume2.1 Studies on Bacteriostasis in vitro and acute toxicity testBacteriostasis test was undertaking to verifying antimicrobic traditional Chinese medicine compound.Twofold dilution was used in this paper to detecting the Minimum Inhibitory concentration (MIC) and minimal bactericidal concentration(MBC) in johnny carp and acute toxicity test was adopted.The result:the MIC to Aeromonas sobria,Edwrdsieda torda and Yersina nuckerij were 3.125mg/ml,0.781mg/ml and 0.781mg/ml; the MBC to Aeromonas sobria,Edwardsieda torda and Yersina nuckerij were 12.500mg/ml,3.125mg/ml and 1.563mg/ml;the median lethal concertration(LC50) for 48 hours was 625 mg/kg; the safe concentration was 62.5mg/kg. It was verified that the Fructus Mume compound was effective for bacillary septicemia treatment.2.2 Studies on pharmacodynamics of Compounds Fructus MumeArtificial conteracting toxic substances were adopted,Throug Liver functional parameters,antioxygen parameters mortalities and medical protections,the effection of Compounds Fructus Mume were reflected. The result indicated: Compounds Fructus Mume with the functions of degrading contents of glutamic-pyruvic transaminase, glutamic-oxal(o)acetic transaminase, bilirubin total,protecting liver cell, strengthenning Liver function, etc. Compounds Fructus Mume also be able to cleanning free radical up in vivo, strengthenning superoxide dismutase activity, protecting cells be injered, degrading malonaldehyde, etc.The Compounds not only be able to prevent and cure bacteriosis in fish,but also be able to degrading the fish with disease mortality. 1% medical protecting rate was 47.73%.
