In this study,using Agilent column(250mm×4.6mm,5μm),methanol - water gradient elution as mobile phase to study the Chinese medicine Fructus Arctii, at the same time in burdock Arctiin and Arctigenin are determined.Arctiin in burdock has a good linear relationship in the scope of 0.005mg/ml-0.025mg/ml (r = 0.9998),and Arctigenin in the scope of 4.64×10-4mg/ml-2.32×10-3mg/ml (r = 0.9996) has a good linear relationship.In the above-mentioned chromatography conditions,FructusArctii of 57 different areas collected from entire country are measured for the content of Arctiin and Arctigenin,the results show that the method is simple,accurate and specific.And carrying out the HPLC fingerprints analysis of Arctii,for accurate and comprehensive evaluation of the quality of medicine to provide qualitative and quantitative methods.Through the content of cluster analysis,cluster analysis of fingerprints and similarity show that the quality of the three provinces in the north-east of China are the best.Stop coughing and eliminate phlegm with 10 different polarity solvent extracts to obtain pharmacological data;and HPLC analysis is developed to obtain the chemical data of 40 peaks of extracts with water and different solvents. Correlation analysis and regression analysis are applied to interrelate the chemical data and pharmacological data.The therapeutic material basis is defined through the effects of stopping cough and expectorant in Fructus hrctii.Studying the chemical constituents of serum fingerprinting of different polarity solvent extracts of Fructus Arctii shows that administrating the same dose of ethanol extraction,ethyl acetate extraction,acetone extraction,water extra- ction and petroleum ether extraction,the content of Arctiin and hrctigenin in the serum of water extraction are more higher than the others, suggesting that other constituents in water extraction may have a promotion role. Pharmacodynamics methods for therapeutic material basis of stopping cough in Fructus Arctii with water extraction is established.The pharmacodynamics process of cough frequency and incubation period indicate that the cough incubation period prolonged and the cough frequency decreased after administration of water extraction.Pharmacodynamics in the process of dose,4 times the amount for cough frequency and incubation period has the best effect.A simple and sensitive HPLC method is developed for the determination of arctiin and arctigenin in serum and applied for its pharmacokinetic study in mice after administration of Fructus Arctii extracts with water.The plasma samples are analyzed with external reference method.The linear ranges for arctiin and arctigenin in rat plasma are 0.029-0.089mg/ml and 0.025-0.044mg/ml respectively.At the same time,seting up HPLC methods measuring arctiin and arctigenin content in the organs of mice,arctiin is the highest levels in lung tissue,and arctigenin is the lowest level in lung,indicating that arctiin is better than arctigenin in stopping coughing role.At the guide of the theory and clinical practice of TCM,the therapeutic material basis is elucidated in this paper.Pharmacokinetic studies of arctiin and arctigenin are carried out in mice to take a limited view of pharmacokinetic profiles of Fructus Arctii.This research provides a significant exploration for the modernization of Chinese traditional medicine.
