In this study,the polymorphisms of 10 candidate genes including FSHβ,ESR,PRLR,BF, Lep,EGF,RBP4,OPN,BMP15 and FUT1 in Beijing Black pigs were analyzed by PCRRFLP or direct electrophoresis,and association analysis of their polymorphisms with reproductive traits(Total number born,TNB;Number born alive,NBA;Birth weight,WB) were analyzed.The results as follows:The PCR products of FSHβfor locus 2,ESR,PRLR,BF,LEP,RBP4,BMP15 and FUT1 were digested with restriction enzyme which were HaeⅢ,PvuⅡ,AluⅠ,SmaⅠ,HinfⅠ, MspⅠ,BciT130Ⅰand HhaⅠ,respectively.While polymorphisms of PCR products for EGF, OPN and FSHβfor locus 1 were detected with direct electrophoresis.The results showed that two alleles and three genotypes of FSHβfor both loci,ESR,PRLR,BF,Lep,RBP4,and FUT1 gene,while AA and AB genotype of BMP15 gene,were detected in Beijing Black Pigs. Two alleles and three genotypes were also detected in FSHβfor locus 1 and EGF gene deduced by deletion mutation of 274bp in intron 1 and 786bp in intron 3,respectively.No polymorphism was detected in OPN gene because only BB genotype was appeared in Beijing Black Pigs.Values of polymorphism information contents,effective number of alleles showed that the mutations of FSHβfor both loci,BF,EGF and BMP15 gene in Beijing Black Pigs were in low polymorphism,while ESR,PRLR,LEP,RBP4 and FUT1 gene in moderate polymorphism. The results of X~2-test showed that all mutations fitted in Hardy-Weinberg equilibrium (P>0.05).Association analysis of polymorphism with litter size indicated sows with AA genotype had larger TNB and NBA than AB and BB genotype in two loci of FSHβgene(first parity P<0.05,later parity P>0.05).Sows with AA and AB genotypes had larger TNB and NBA than BB genotype in PvuⅡlocus of ESR gene(P<0.05),and allele A had positive effective on interest traits.Sows with AA genotype had larger TNB and NBA than AB and BB genotype in AluⅠlocus of PRLR gene,which was significant difference in later parity but not in first parity.Sows with AB genotype had significantly larger TNB and NBA than AA and BB genotype in SmaⅠlocus of BF gene in later parity.No significant differences in sows with different genotypes of Lep and EGF gene were found on litter size.Sows with BB genotype had larger TNB and NBA than AA and AB genotype in MspⅠlocus of RBP4 gene(first parity P>0.05,later parity P<0.05).Sows with AA genotype had larger TNB and NBA than AB genotype in BciT130Ⅰlocus of BMP15 gene,and the difference was statistically non-significant(P>0.05).Sows with AA genotype had significantly larger TNB than that with GG genotype in HhaⅠlocus of FUT1 gene in later parity(P<0.05).Six loci in five genes including 274bp deletion mutation and HaeⅢlocus in FSHβgene, PvuⅡlocus in ESR gene,AluⅠlocus in PRLR gene,SamⅠlocus in BF gene,MspⅠlocus of RBP4 gene and HhaⅠlocus of FUT1 gene,maybe associate with reproduction traits in Beijing Black Pigs,and increasing genetic progress of productivity could be obtained by using MAS.
