Wolbachia is a kind of maternal inheritance intracellular symbiotic bacteria existing in the arthropod, which can be transmited through cytoplasm by eggs and involved in some reproductive activity of host, such as parthenogenesis inducing, cytoplasmic incompatibility and so on. Wolbahia may be a kind of symbiotic bacteria that distributes most widely and owns the largest abundance. Wsp gene encoding the surface protein of Wolbachia and it evolved much faster and can reveal the interrelation between Wolbachia better as compared with 16SrDNA,ftsZ and other DNA section.Tetranychus urticae Koch distributes and host widely. Because of the mass and inproper use of pesticide in fruit zones of Shaanxi in recently years, the resistance makes it become the main pest on fruit, and bring more damages than the other crop pest. Recently research shows that the parthenogenesis may closely relate to the endosomatic symbiotic bacteria. Therefore, it may be a way to control the Tetranychus much more effectively that induces the abnormal reproductive by symbiotic bacteria Wolbachia, such as parthenogenesis, masculinization and so on, which can be said using the population with less damage to replace the original population.With the advocation of the prtoducational technology on apples with pollution-free and high quality in recent years, use symbiotic bacteria Wolbachia to control Tetranychus urticae Koch is a feasible and exact way. Use universal primers for wsp gene of Wolbachia and Polymerase chain reaction (PRC), we investigated and cloned the Wolbachia infection of four populations of Tetranychus urticae Koch on four different host plants in Shaanxi province. Meanwhile, we compared the homology and analyzed the gene phylogeny of the sequenced PCR products under Phylogenetic Tree Observed Divergency of DNAMEN 6.0. Results showed that all the populations were infected, and the region of 596 bp of Wolbachia wsp gene were amplified from the four populations;By analyzing the wsp gene sequences of Wolbachia and constituting the phylogenetic tree, the strains of Wolbachia, which infected Tetranychus urticae Koch ton the Malus pumila and Prunus persica had the highest homology of 100%, while, Tetranychus urticae Koch that on the Malus pumila and Viola vaginata Maximhad the homology of of 99.2%. They were similar to or completely identical with the wsp gene sequences of some strains of Wolbachia among Ori group in B supergroup. Since the branches of clusters are parallel, there is no evident host plants distribution clustering between different populations.
