Inonotus obliquus belongs to Eumycota, Basidiomycotina, Hymenomy, Aphyllophorales, Polyporaceae, Poria. It grows in the Frigid Zone. It is the wood rot fungi.In this paper, we used the sclerotiun of Inonotus obliquus as the material,and used distilled water as the extraction agent. The water soluble crude polysaccharide was extracted by boiling water. Then we can obtain Alkali polysaccharide with the method of Alcohol Precipitation, Dialysis and dry from the Residue. The Average recovery rate of water soluble crude polysaccharide is 6.7%. Sugar content is 23.5%. The Average recovery rate of Alkali polysaccharide is 4.7%. Sugar content is 14.8%. Through the method of TLC and GC, we know the water soluble crude polysaccharide main contain glucose, mannose, galactose, the Arab sugar and the Most of the content is glucose(34.4%).we also know the Alkali polysaccharide main contain glucose, mannose, galactose, the Arab sugar and Rhamnose.In order to know the distribution of molecular mass of the extracted crude polysaccharide, we used the Sepharose CL-6B gel exclusion chromatography to find the molecular mass distributed Wider. From the experiment of soluble polysaccharide stimulating Lymphocyte proliferation, we know 30μg/ml is the best Concentration to stimulate lymphocyte proliferation and it possesses the function of working together with ConA and LPS. We used the method of MTT to detect the Anti-tumor activity of polysaccharide, then we find that when Concentration is 200μg/m, the Inhibitory rate of the water soluble crude polysaccharide to Hela is 57.7%.
