Carnation Fusarium wilt disease which caused by Fusarium oxysporum is a kind of soil-borne diseases.This disease is distributed in all of the world carnation productive areas,as well as widely happened in planting areas in China which is serious harmful and contributes to huge economic losses,it becomes a major obstacle factor of carnation high yield and good quality.Bioteehnology methods were used to integrate dual-price resistance gene GAFP-NPI into carnation,foster transgenie plants,the past researches demonstrated that transgenic carnation have antiacterial activity on Trichoderma viride.This study isolated pathogens from the carnation Fusarium wilt disease,then utilized different methods to measure transgenic carnation anti-Fu sarium wilt disease resistance,following are the results:1.The tissue culture experiments revealed that transgenic carnation varieties are more resistant than receptor carnation varieties,mainly because the transgenic carnation needs much more time to wilt than receptor carnation.2.Through in vitro seedlings to pathogens found that when compared with receptor carnation,the resistance of transgenie carnation had been noticeably improved,which displayed in the length of lesion expansion of transgenie carnation is shorter than receptor carnation.3.Determined the best conditions for Fusarium oxysporum producing toxin, with different concentrations of toxins from the body deal with carnation seedlings, found that 50%concentration of the toxin shows the best identification effect. Utilize 50%toxic liquid handling to transgenic carnation and non-transgenic carnation,the resistance of transgenic carnation improved markedly.4.Established a carnation suspension cell lines.With Fusarium oxysporum toxic liquid handling of transgenic and non-transgenic carnation Carnation suspenension cells,reached the following conclusions:interaction after the Carnation transgenic cells mortality rate lower than that of transgenic carnation;the cell death of the transgenic and non-transgenic carnation have little difference;transgenic carnation relative permeability of the membrane is lower than that in transgenic carnation.
