the genetics of O/L in F1 and backcross population were also studied. The result were:1. In F1 population, the mean O/L ratio was higher than that in their parents, indicative of the existance of heterosis. In F2 population, an average of 0.133 increass in O/L ratio was observed. The result is that F2 may be used.2. Comparation of O/L ratio in F1, P1 -BC1 and P2-BC1 populations. To understand the genetic characters, the F1 plants were backcrossed with P1 (low O/L) and P2(high O/L) plants, in P1-BC1, the O/L ratio of most plants were fell into 0~1.2 and the highest rate was 3.52.In P2-BC1 population, more seeds tended to be with higher O/L and the highest rate was 38.94. From F1 to P1-BC1 and to P2-BC1, O/L showed an upward tendency, and the average approached to the regression parent.In a parallel work in AFLPResearched purpose: offering transformed chance from difficult AFLP to simple SCAR , Studying the function of fatty-acid in raising peanut fatty acid and offering reference of planting fine peanut. Researched way: A peanut variety Sanyou 162(P1) with a low O/L was crossed with Sunolic95R with high O/L(P2) and the F1 population were used for genetic annlysis with AFLP marker. The results were as follows:3. One hundred and fourteen of EcoR I /Mse I primer pairs were used to amplify the two parent peanuts and 77 of them detected 170 polymorphic bands, the polymorphic frequency was 2.8%.4. In studying restriction-ligation of genomicDNA, separately doing is better way than two steps and one step. Gene parts can meet 100 bp~1800 bp. Dilute and no dilute ligation produces affect preamplifi cation, the result depend on DNA sum in restriction. Dilute and no dilute preamplification prod uces affect selective ampli -fication, the result is that diluting 20 times is the best.5 . One hundred and fourty-five polymorphic AFLP bands were recovered and twenty of them were cloned and sequenced. The sequences were compared by blastn anc blastx in the GenBank and the result is that no identity, the further work is in progress.
