Castor(Ricinus conununis L.)is dicotyledonous plant,one of the world's top ten oil crops.Castor oil is mainly composed of ricinoleic acid and other ingredients.2129 is an important breeding male material castor in Tongliao area.Methylation-sensitive amplified polymorphism(MSAP)technique,cDNA-AFLP technique successfully used to study the genetic mechanisms of gene expression,linkage mapping,functional marker analysis.In this study,we used strain 2129 for research,measured different developmental time hundred fresh grain castor seeds absolute content of various fatty acids,using cDNA-AFLP and MSAP markers technique to study fatty acid content in differences development time castor seeds gene expression screening different bands,and sequenced,functional annotation,in order to lay the foundation for improving the quality and breeding of high castor oil.Main results are as follows:1.Complete determine 2129 of six different developmental time castor seeds fat and fat in various fatty acids(ricinoleic acid,linolenic acid,palmitic acid,stearic acid,arachidic acid,linoleic acid,peanut acid)the absolute content;2.MSAP technology system analysis differencent gene methylation level of 2129 seeds at different developmental time.Total of 32 primer combinations used for six different developmental time castor seeds differentially expressed gene amplification and obtained 647 significantly differentially expressed genes bands.Through NCBI homology retrieved 37 differentially expressed genes with known sequences high-derived.Homologous protein gene expression include eight major categories,including FAD oxidoreductase(B02),ATP combined with transport enzyme(195),?-galactosidase/?-positive acetyl aminotransferase(D16),peroxide enzyme(F13),they direct or indirect relationship with the fatty acid synthesis,transport.3.cDNA-AFLP technology system used for separate and analysis 2129 differentially expressed genes at different developmental time seed.62 pairs primers for differential gene amplification,to give 534 differentially expressed genes,26 sequence homology function through NCBI database search finds genes with known function of high homology.Homologous protein comprises 10 categories,including acetyl coenzyme A carboxylase(177),fatty acid synthase(108),fatty acid desaturase(48),acyl synthase(87)with a fatty acid synthesis,transport,storage and other physiological biochemical processes directly or indirectly.
