Abstract/Summary:
Many profitable genes are contained in wild rices which are important gene resource extension of dominated rice. Traditional hybrid methods propose the limits of reproductive isolation and difficulties of cloning directly from wild rice because of poor heredity background. The research comprehends the candidate resistance gene cloning strategies, utilization the ability of RecA enrichment library and the transformation-competent genomic library technology. The research includes the construction of wild rice transformation-competent genomic library and the RecA protein mediated biotinylated probes enrichment library. And then, point hybrid and the terminal sequencing are taken before the whole sequencing and function analysis. The research accomplishes the Dongxiang and Xiaoli wild rice genomic libraries which cast 97.10% and 98.42% of the whole genome. Twenty-two positive clones are isolated and the terminal sequencing reveals that some of the clones contain whole new resistance genes. The result suggests that the strategy of cloning new resistance genes by constructing enrichment genomic is feasible.
