Many favorable genes are contained in wild rice .Traditional crossing methods and the cloning strategies of resistance genes commonly used are difficult to be used because of reproductive isolation and poor heredity background. Our research comprehends the RecA enrichment library and RGAs(resistance gene analogs) strategies to screen Oryza minuta pCAMBIA1301 liquid genomic library which contains many resistance genes. 11 STK (Serine/threonine kinase) positive clones were obtained. 4 of them are located in the near areas of resistance genes or the resistance gene itself by the terminal sequencing analysis. 3 of them can not be mapped in Japonica or Indica genome. Probably they are the special sequence of Oryza minuta. 55 NBS (Nucleotide binding site) positive clones were obtained. 27 of them were analysised and 1 positive clones can be located to the resistance genes. Some can be located to the areas which were not clearly researched. And the others can not be located to Japonica or Indica genome. But they have high matching with Oryza minuta and Oryza australiensis. And then, RPS2 positive resistance gene was amplified by LA-PCR. The analysis indicated that it has deletions in LRR motif. The identification for the resistance to rice bacterial leaf blight indicated that it was susceptible to rice bacterial leaf blight ,which could be because of the deletion of LRR motif. Other 526 transgenic rice plants were identified for the resistance to rice bacterial leaf blight. The results indicated that 7 (6 class positive clones)of them were medially resistant to rice bacterial leaf blight.
