The paper was studied on Mucor M-1 to the safety of the mice,growth amount and coarse protein content in the liquid medium and the fermentation media,optimization of the fermentation condition using soybean meal as the main raw material,effect of fermented soybean meal and Bacillus PAS38 on the growth of rabbits.1.The collected spore suspension of Mucor M-1(amount of spore was 3×1011cfu/ml)and fermented stock solution were given 0.5ml dose to the average weight 18g mice by intraperitoneal injection,observed for 7 days and found no abnormalities.The result showed that Mucor M-1 was relatively safe.2.Mucor M-1 could grow in the medium lacking of nitrogen sources,but the growth is poor,the results conjectured that Mucor M-1 had certain nitrogen fixation.It could grow better in the medium contains trace nitrogen(0.025%of yeast extract) and Sandburg medium,it showed that Mucor M-1 could grow in trac nitrogen medium.The crude protein content ofMucor M-1 reached 57.67%while Mucor M-1 growed in Sandburg medium.3.The protease activity of cellulase and amylase produced by Mucor M-1 in the fermentation medium was low and could not be detected.The results suggested that Mucor M-1 in the fermentation medium need little protein,even didn't need protein.Coarse protein content of soybean meal which ferment by Mucor M-1 increased 11.49%than the control group.It showed that Mucor M-1 was a good strain for producing biological protein diet by fermentation.4.For choosing the optimal fermentation condition(including cellulose enzyme, amylase and matrix coarse protein content)ofMucor M-1,the five factors(ratio of material to water,the temperature of fermentation,fermentation time,inoculation amount,the initial medium pH) which could influence the fermentation under different conditions was detected after the Mucor M-1's fermentation medium was determined.The results showed that the best fermentation conditions for cellulose enzyme were:ratio of material to water 1:1.5,temperature at 30℃,fermentation time for 72 hours,the inoculation amout 20%,the initial pH 5.5.The best fermentation culture conditions for amylase were the same as cellulose enzyme's,but the fermentation time was 84h.The best fermentation culture conditions for matrix coarse protein content were the same as cellulose enzyme's.5.30 weaned rabbits were randomly divided into 3 groups for feeding test.We used fermented soybean meal in basic diet(group 1),added Bacillus PAS38 in basic diet(group 2) and only basic diet(control group).30 days after the feeding,the body weight,the feed conversion,the immune organs index,the cellulose enzyme,protease and amylase activity of jejunum and the cecum and the number of live bacteria in rectum were calculated.The result showed that the body weight of the experimental groups was significantly higher than control group,and the goup 2 was better.Compared to the control group,the weight and organ index of thymus and spleen of the experimental groups showed extremely significant increase.The weight of sacculus rotundus of the experimental groups showed extremely significant difference,and the difference in organ index was significant.The weight of vermiform appendix of groupl was significant increase,and the group 2 was extremely significant,but the organ index of the experimental groups was not significant.The number of E.coli,Aureus and Anterococcus in rabbit rectum content decreased,the difference was significant in the number of E.coli of group 1,E.coli of group 2 was extremely significant,Aureus of group 2 was significant,but,Aureus of group 1 and Anterococcus the experimental groups were not significant.The number of Lactobacillus and Bifidobacterium of experimental groups in rabbit rectum content increased significantly.The activity of cellulose,amylase and protease in experimental groups were higher than in the control group.Among two experimental groups,the differences of cellulose and amylase were extremely significant as the difference of protease was not significant.
