Common buckwheat is the key cultivated buckwheat in genus Fagopyrum Mill (Polygonaceae). Common buckwheat and its 17 trisomic lines were studied by means of real-time PCR, cytology and morphology. The main results were as follows:1. It is the first time to set up the identification system of common buckwheat trisomic lines by means of the real-time PCR.2. The real-time PCR analysis of the trisomic lines of common buckwheat showed that: the lines T13 and T14 are the same trisomic line identified by Primer 26; The lines T4 and T7 are the same trisomic line identified by Primer 32; The lines T2, T6, T10, T16 and T17 are the same trisomic line identified by Primer 27; The lines T9, T11 and T12 are the same trisomic line identified by Primer 33. These results indicated that the amplificative products of Primer 26, Primer 32, Primer 27, and Primer 33 locate in different chromosomes of common buckwheat, respectively.3. The karyotype analysis of common buckwheat root tips showed that common buckwheat is diploid, 2n=2x=16, and has two pairs of satellite chromosomes located in 4E and 8E chromosomes, respectively. Its karyotype type is 1A. All chromosomes have the arm ratio of 1.01-1.70 and are m chromosomes. Its karyotype formula is 2n=2x=16=12m+4m(SAT).4. Karytype analysis of trisomic root tips also showed that: the lines T2, T6,T10 and T17 are the 1E chromosome trisomic; T1 is the 2E chromosome trisomic; T13 and T14 are the 3E chromosome trisomic; T8 and T15 are the 4E chromosome trisomic; T3 is the 5E chromosome trisomic; T4 and T7 are the 7E chromosome trisomic; T9,T11 and T12 are the 8E chromosome trisomic.5. Real-time PCR and karytype analysis of trisomic showed that the primer 27, primer 26, primer 32, and primer 33 can identify 1E, 3E, 7E, and 8E trisomics, respectively.6. Morphological analysis showed that there are some differences among the trisomic lines of common buckwheat, especially 3E, 4E, 5E, 6E, and 8E trisomics. 3E trisomic has higher plant, more flowers, and 4 or 5 splits of style. 4E trisomic plants are dwarfer, more compact, more branches, shorter branches and knotes, and more leaves; 5E trisomic plants are weaker with medium height and have spindly stems, fewer leaves, shorter anthesis, and lower seed set; 6E trisomic plants are much weaker and produce more two-arris achenes than others. 8E trisomic plants have longer branch and knotes, slighter and longer branches, and their perianths are usually more than 5.
