HG pectin was prepared from the crude water-soluble extraction of the roots of Panax ginseng C. A. Meyer by partial hydrolysis. The carbohydrate content of GR and GRP was 57.90% and 99.84%, determining by the method of phenol-sulfuric acid. The uronic acid content of GR and GRP was 35.30% and 97.31 % according to the method of m-hydroxyphenyl. The result of HPLC showed that GRP was composed of GalUA with trace Glc, Ara and Gal. GRP was mainly consisted ofα-(1→4)-D-GalUA with branch at position 2 or 3 by the analysis of pectinase hydrolysis and 13C NMR.Using the pectinex ultra SP-L(novozymes)to hydrolyze the GRP, breakingα-(1→4)-D -GalUAP, separated with Sephadex G-75 , we obtain the non-α-(1→4)-D–GalUA structure fine unit of GRP, enhance the saccharide detecting sensitivity by pre-column derivation, obtain the partial non-α-(1→4)-D–GalUA structure fine unit by the HPLC ODS column, carry on the preliminary analysis through the monosaccharide composition determination of its fine unit.
