The genetic diversity of eighty-one Galege L. germplasm materials introduced from Russia was analyzed by phenotype diversity and ISSR \ SRAP marker.Main result obtained are as follows:1. The result of phenotype diversity shows that the Galege L. plants rich in morphological variation, the average coefficient of variation is 25.99%. Based on the morphological data, all materials were clustered into four groups at genetic distance of 7.76. The findings implied a strong correlation among the germplasm materials and geographical environment.The materials from the same origin were clustered into one group. The average of Shannon-Weaver index of G. officinalis L.and G. orientalis L. was 1.863, 1.913, respectively. The results showed that the genetic diversity of Galege L. germplasms was abundant.2. Inter-simple sequence repeats (ISSR) and Sequence-related amplified polymorphism (SRAP) marker amplification system were optimized. ISSR-PCR was performed in a 25μl reaction mixture with 20ng DNA, 2.0mmol/L Mg2+, 0.2mmol/L of each dNTP, 0.2μm/L primer, 1U Taq polymerase and 1×Buffer. SRAP-PCR was performed in a 25μl reaction mixture with 20ng DNA, 2.0mmol/L Mg2+, 0.2mmol/L of each dNTP, 0.3μm/L of each primer,1U Taq polymerase and 1×Buffer.3. The average polymorphic ratio of ISSR and SRAP marker of G.officinalis L was 76.04%, 65.52%, respectively. The result showed that there was rich genetic diversity with the G. officinalis L. The Nei's genetic similarity coefficient of the germplasm materials ranged from 0.6 to 0.8, and the average Nei's coefficient was 0.66,0.688, respectively. The result of genetic similarity (GS) analysis showed that the result of GS value of two molecular marker were consistent. Based on the ISSR and SRAP marker, the thirty-five materials of G.officinalis L. were clustered into four groups at the GS (0.70). The materials from the same origin were frequently clustered into one group. The findings implied a strong correlation among the germplasm resources, geographical and ecological environment.4. The. average polymorphic ratio of ISSR marker of G. orientalis L was 78.65%. The results showed that the genetic diversity with G. orientalis L. was abundant. The Nei's genetic similarity coefficient of the germplasm materials ranged from 0.32 to 0.98, and the average Nei's coefficient was 0.58. Abundant genetic diversity with G. orientalis L. materials could be revealed by ISSR marker. Based on the ISSR marker, the thirty-eight materials of G. orientalis L were clustered into five groups at the GS (0.59). The materials from the same origin were frequently clustered into one group. The findings implied a strong correlationwith the germplasm resources, geographical and ecological environment.
