The apricot (prunus armenniaca L.) belongs to the stone fruit , China is one of primary ceter of apricot domestication in the word, owning very abundant germplasm resources of apricot.The traditional methods, morphology marker and palynolony,has some limitations.In recent yeas, the molecular marker becomes a new identification method. The four leves of five factors (Mg2+, dNTPs, primers, Taq DNA polymerases ,template DNA ) in ISSR-PCR systerm was studied by L1(645)Orthogonal- design ,and then establish and optmize the ISSR-PCR reaction systerm of Kernelled Apricot for the future apricot breeding .For the first time using ISSRmarker combined with morphological methord to research 39 apricot germplasm.,the purpose is to reveal the diversity of germplasm resources in china, aimed at breeding superior new apricot.The same time ,through comparative analysis of two makers to find a moer effective tool for genetic analysis.We get the following conclusion by the experiment:1. Finally establish an optimal reaction system of PCR for Almond-apricot which is: in a 20ul PCR reaction system contains 1.5ul Taq DNA polymerase,2.0mmol/L MgCl2,0.15mmol/L dNTPs,0.35umol/L primers and 30-60ng template DNA. And the best response system is validated. Taq DNA polymerase and Mg2+ are the two main affective to ISSR-PCR reaction.2. With 18 informative primers in ISSR analysis,there are 153 amplified bands in total, 132 polymorphic bands,and polymorphic frequeccy is 0.863.3. The survey of the difference and variation degree of 22 apricot quantitative characters showed that: apricot quantitative characters has more different traits, the biggest variation coefficient is fruit size (84.5%), the smallest is the leaf width (11.9%).4. ISSR markers UPGMA cluster analysis showed that the apricot has four groups:Asia eco-geographical sub-groups,orthern China eco-geographical sub-groups,urope eco-geographical sub-groups,Apricot of Kernel-Consuming (Armenica spp.Mill)group.He varieties showed strong concentration of the geographical distribution of apricot. Within the group,apricot has close affiliation between different species.5.By the 22 apricot quantitative traits cluster,39 apricot germplasm can be clustered 7groups,not match to ISSR marker cluster result .Constrasted with previous studies and this study ,found that ISSR marker can be used a more efficient analysis tool.
