| Peking duck is a famous breed of ducks. It plays important roles in ducks breeding and ducks industry development in the world. But the development of breast muscle of Peking duck has two characteristics. The breast muscle rate was low and the development of breast muscle was relatively late.In order to improve the development of Peking duck's breast muscle,we have researched the relative analysis of the polymorphism candidate gene with the candidate index of breast muscle . The results were as follow:1,Complete MSTN promoter regeion was detected by PCR-SSCP in four lines of 300 Peking duck(sZ2 line,Z4 line,Z4×Z2 Cross line). We found difference haplotypes, and compared with sequence of the register number AY329600. Four SNPs -753,-737,-658,-235 were detected (relative to ATG start codon). These alleles were in linkage.2,It showed two genotypes (MM,MN) at F locus and two genotypes (RR,RS) at D locus in Z2 line and Z4 line of Peking ducks. It showed three genotypes (MM,MN,NN) at F locus and three genotypes(RR,RS,SS) at D locus in Z4×Z2 Cross line of Peking ducks;Individuals do not have NN /SS in the lines of Z4 and Z2.The frequency of allele M/R was dominant in all four lines. The polulatin of Z2 , Z4 and Z4×Z2 were not at Hardy-Weinberg equilibrium (P<0.05).3,Variance analysis showed that MM and RR genotype birds had significant higher weight of abdominal fat percentages (P<0.05 )and breast meat percentages(P<0.10) than MN and RS genotype in the lines of Z2. MM and RR genotype birds had significant higher weight of breast meat percentages(P<0.10) than MN and RS genotype in the lines of Z4. MM and RR genotype birds had significant higher weight of abdominal fat percentages (P<0.05 )and breast meat percentages(P<0.10) than other genotypes in the lines of Z4×Z2 cross line.4,321bp cDNA of MSTN gene 3'UTR was cloned form RNA isolated from Beijing duck's breast muscle and it was detected by PCR-SSCP in four lines of 400 Peking ducks.We didn't find any polymorphism.
