The newest growth part of seeds of reciprocal hybrid F1 between Agropyron mongolicum Keng.(2n=2x=14) and A. cristatum gaertn. cv. Fariway (2n=2x=14) were doubled by colchicines; and the variated plants from above seeds were identified and analysed based on anther features, pollen sterility, chromosome pairing configuration, isozyme zymograms phenotypes and AFLP maker. The main results as follows:1. The suitable colchicine concentration and treatment time for the seeds of reciprocal hybrids F1 between Agropyron mongolicum and A. cristatum cv. fairway were 0.15%~0.20% and 24h~36h respectively, the suitablest colchicine concentration and treatment time in this experiment were 0.20% and 24h respectively, and the average variated rate of the seeds was about 7.6%.2. The chromosome number of varited plants'cells of reciprocal hybrids F1 was 28(2n=4x=28); the bivalent frequency of chromosome doubling plants of reciprocal hybrids F1 were nearly 14 at metaphaseⅠin pollen mother cell, the chromosome pairing configuration was more regular, all of those was cytology basis about fertility restored.3. The anthers of chromosome doubling plants of reciprocal hybrids F1 of Agropyron mongolicum and A. cristatum cv. Fairway were larger, the surface ornamentation of anther wall arranged regular, the content matter was plump, so anthers was normal growth; its pollen fertility rate were 78.16% and 79.29% respectively, and was improved to the level of their parents.4. There were certain differences in the number, locus and intensity of the proxidase (POD) and esterase (EST) isozyme bands among the reciprocal hybrids F1 and its chromosome doubling plant and their parents at tillering and earing stages.The differences of isozyme zymograms phenotypes were important genetic markers in protein level to the reciprocal hybrids F1 and its chromosome doubling plant and their parents.5. The genomic DNA amplifing of AFLP markers with 11 paris primers to the reciprocal hybrids F1 and its chromosome doubling plants and its parents gained 430 bands, their percentage of polymorphism loci was 92.10%, and DNA fragments were range from 100 to 1500bp. the materials experimented on were significantly different from each other in AFLP fingerprint, and the fingerprint can be used as molecular marker to identify every one of them.The above results can be used as theory foundation for breeding new cultivars of crested wheatgrass.
