On the basic of some research aboutβ-GAL gene family, 5' flanking sequence and the promoter ofβ-GAL homologous gene were isolated from papaya genomic DNA via three times chromosome walking of IPCR, and then the function of the promoter was identified. All of these established a foundation of the reaserch on everyβ-GAL gene's effection on papaya fruits'softening via RNAi technology in promoter region to prolong the fruits'shelf life. The main results were as follows:1. 5' flanking sequence and the promoter ofβ-GAL homologous gene were isolated from papaya genomic DNA, and the promoter was analysed by software online.5' flanking sequence and the promoter ofβ-GAL homologous gene were isolated from papaya genomic DNA by IPCR, and then submitted to GenBank, whose accession number was EF694758. The core promoter region and transcriptional start site(TSS) were predicted using Neural Netwokr Promoter Prediction software online. The result was that core promoter region maybe existed at the region 195-245bp, 552-602bp, 560-610bp and 591-641bp. Plant CARE and PLACE software were also used, the promoter region we predicted had some regulated motif related to fruits' ripenness and some impotant elements also in other plants' promoter, like ABA motif.2. The promoter's function was identified preliminarily.Chimeric vectors contained different length promoters were constructured, and then were transferred into papaya flesh mediated by agrobacterium transformation. The result of transient GUS expreesion indicated thatβ-GAL homologous gene promoter can drive gene expression in papaya flesh. By the time and degree of colouration, it was deduced that there were some negative elements between 603bp and 724bp probably.
