Loquat leaf is one of the most common Chinese traditional medicinal crops.The current reseach topic is mainly focused on the extraction, isolation, purification of loguat leaf poiysacharides(LLPS) and characterization of their chemical properties and biological activities. By means ofsome modern techniques and methods, the following preliminary results were achieved.(1)The results of single factor tests and orthogonal experiments demonstrated that the optimum LLPS extraction conditions are as follows: solid-liquid ratio is 1:15,temperature is 85℃,extraction time is 4 hours, the extraction times is 3 and precipitated with four times of alcohol. Under this condition, the content of total sugar is 56.7 %.(2)After powder of loquat leaf was extracted, LLPS were got by a series of steps, such as eliminating starch, getting rid of protein with Sevag's method, isolating with 95% alcohol and washing with 80% alcohol,and so on. DEAE-cellulose chromatography was used to separate crude LLPS, further purification was carried on by SephadexG-200 chromatography. Finally, it's homogeneity was proved by HPLC.(3)The results of the vitro-anti-oxidation suggested that LLPS has good anti-oxidation action, with the dosage of 3.0 mg/mL the inhibitory rate on hydroxyl free radical was 69.80 % ,each dosage can inhibite superoxide anion with the dosage of 1.5mg/mL the inhibitory rate on inhibite superoxide anion was 58.12 % ,with the dosage of 5.0 mg/mL the inhibitory rate on the restraining auto-oxididation of erythrocytes was 58.60%,with the dosage of 5 mg/mL the inhibitory rate on lipidperoxidation was 63.1%.(4)The loquat leaf polysaccharide indicated to the molecular material protective function result that LLPS I to the DNA chain, the basic group and the DNA ribodesose all has certain protective function. The protective function is higher than the mannitol slightly.(5)The results of LLPS to immunostimulating activity and antioxidant activity of mice , Mice were fed with formulated diet supplement with 400 mg/kg, 600 nag/kg, 800 mg/kg LLPS running for 10ds. Immunity indexes (IgA,IgG,IgM,C3,AKP,ACP) and antioxidant indexes (MDA,SOD,GSH-PX, T-AOC) are analyzed on the day of 11th respectively. The results showed that LLPS enhanced the immunity and antioxidation of mice significant compared with control group (p<0.05).Diets supplementation with different LLPS no significant effects were observed. In conclusion, LLPS could improve immunity function and antioxidant ability of mice.(6)LLPS I was analyzed by thin layer chromatography (TLC), element analyzer, IR, HPIC, GC. The results show that The results of GC and TLC analysis showed the monosaccharide of is Man,Glc.
