A water soluble crude polysaccharide(EP) extracted by 0.5mol/LNaOH has been isolated from the residue of Enteromorpha prolifera which had been dealt with alchol and hot water . Gas chromatography (GC) showed that it (EP) is a mixture of neutral heteropolysaccharide that composed of Rha, Xyl, Gal, Glc and little Man, GluA, Ara. After removed protein by the combined deprotionization methods of enzyme and sevage and fractionated by ultra filtration and gel chromatography, the polysaccharide (EP2) was obtained. Homogeneity and molecular weight of RCP were determined by Gel filtration on Sephadex G-6B, specific optical rotation measurement and High-performance liquid chromatography (HPLC) analysis. The sugar composition was determined by methanolysis and gas chromatography (GC). The results revealed that EP2 is homogeneous on both molecular weight and polarity. The molar weight is estimated to be 4.6kd by means of HPLC. It was consisted of Rha(12.27), Xyl(1.88), Gal(1.00) and Glc(1.63). EP2 is a neutral heteropolysaccharide.Partial hydrolysis with acid,Periodate oxidation,Smith degradation, I.R.,and Methylation were used on the structure analysis of EP2.After acetylation, the methylated productions were analyzed by G.C.- M.S.Through IR, GC, partial hydrolysis with acid, periodate oxidation, smith degradation, methylation and GC-MS analysis, the result indicate that EP2 has a kind of lowly branched structure. The main chain is mainly made up of major(1→4)rhamnose and some(1→2)rhamnose and (1→6)galactose; The side chain is composed of (1→3) rhamnose, (1→4) glucose, (1→4) xylose and (1→3)galactose; The nonreduced end is composed of rhamnose and glucose. Residues are substituted at 2-O of rhamnose (1→2,4 ) and xylose (1→2,3), On average, there are about 3 branches among 20 residues.A colorimetric tetrazolium MTT method was used for the determination of anti-tumor activity evaluation, the effect is significant. EP and EP2 showed antitumor activity evidently.
