Utilization of heterosis in rapeseed is an important way to increase yield.Male sterility has been widely used in the heterosis breeding program in rapeseed. And development of male sterility lines and restore lines plays an significant role in utilization of cytoplasmic male sterility in rapeseed.Since spontaneous male sterile plants were found in Polima cytoplasmic male sterility(Pol CMS) by Fu in 1972,Pol CMS has become the most valuable CMS in application. In order to make full use of this CMS and improve the yield, quality and resistance of hybrid, developing a elite male sterile line, fine economic and agronomic traits and high combining ability, has been a crux in hybrid rapeseed production.A novel Temperature-sensitive CMS line was selected by Yang in 1990,which had then triggered a booming use of Pol CMS in oilseed breeding. In this research, the possibility of developing an elite TCMS sterile line by high-effective embryogenesis from Yu115,a Pol TCMS varieties, has been investigated.Genetic diversity of the DH lines from Yu-155 was analysed by SSR and AFLP.And 40 SSR primers and 64 AFLP primers were used to analyse the genetic diversity of 72 DH lines.6 SSR polymorphic bands and 220 AFLP polymorphic band were used for clustering analysis with UPGMA.And the results demonstrated the genetic variation among the DH lines was relatively visible. The range of similarity coefficient(SC) in the DH line population varied between 0.57-0.98.If classified at the 0.68 SC level, the DH population could be divided into 3 subgroups.Accouding to both sterility traits and quality traits,11 DH lines, including 115-2-1,115-3,115-6-1,115-11-2,115-23-1,115-92-1,115-94-2,115-95-1,115-99-3,115-102-1 and 115-107-1,were considered as the candidates of sterile line for breeding.
