Inonotus obliquus(Fr.)Pil(?)t is a kind of rare medicinal fungi.On the taxology it belongs to the Basidiomycotina,Hymenomycetes,Aphyllophorales,Polyporaceae,Poria hypobrunnea Petch.It mainly distributes in the area of the Northern Hemisp-herenorth latitude 45°~50°.Sclerotium has the properies of medicine.The amylose of the Inonotus obliquus is one of the mainly medicinal component,it has the effect of restraining the cell cycle and reduce the blood sugar.Recetly due to a great deal pluck and medicine empolder,the wild resource hardly satisfy the need of the market and the amylose of the wild Inonotus obliquu only 1.8%.It is a urgent affair how to cultivate the higher amylose and higher sclerotium production.This experiment uses the Inonotus obliquus stains JL01 as the material which treatmented with ultraviolet and irradiation of 60Co-γ,screen out good productive stains according to the content of amylose.Identity the orrigen stains and good mutagenic stains by the POD and RAPD.The results showed that the strains irradiated 20min and 25min for 3 times are the high production and polysaccharide strains and they are named JLZ1,JLZ2;the strain of the polysaccharide content of Inonotus obliquus mycelium reach the maximum value when the 60Co-γirradiation is 1000 Gy and irradiation rate is 48.31Gy/min was named JLC1;the result on the peroxidase isozyme indicates that there are 6 common bands in JLZ1,JLZ2,JLC1 and the number and expression location were very alike except the colour of bands,all this indicates that the gene of mutagenic stains has changed;RAPD amplification result indicate that six random primers were amplified with 74 bands,of which polymorphic bands are 28.Six random primers all show characteristic bands that mutation strains is different from the original strain,this showed that gene of mutation strains irradiated by UV and 60Co-γhave changed from origin strain.RAPD results are consistent with isozyme and JLZ1,JLZ2,JLC1 can be founded to be mutation strains...
