The mose rose(Portulaca grandiflora) is an annual herb flower which belongs to Portulaca Linna,and a fine traditional cultured of summer flower.Now it is widely used in gardens and family prettification.But at present many of varieties sought in the market are imported from overseas and the price is too high.Therefore,the cultivation of new variety of mose rose which has the proprietary intellectual property rights is urgently awaits to be solved at present.In this study,three S1 generations,one F1 generation and seven M2 generations of mose rose were used to obtain pure inbreeding lines by pedigree method;polyploid species were gained by treating mose rose seeds and young plants with colchicine and these species can be used as interspecies cross materials.The main results were as following:Selection of self-bred lines:â‘ Twelve self-bred lines which have consistent characters in shape of stem and flower color were obtained by pedigree method, including two S3 generation,five S2 generation and five M4 generation.â‘¡Different series seed germination ratios and pollen germination ratios were observed and analyzed: under the identical germination environment,six radiate M3 generation lines germination percentage were higher than S2 series obviously,in the inbred line series,within M3,S2 inbred lines,germination percentages were similar;Under 0.1%boric acid +5%sucrose +0.6%agar combination,the pollen germination ratio was highest,it was showed that the mose rose pollen germination need the high concentration boric acid and the low concentration sucrose;At different time such as7:30,9:30,11:30,germination ratios did not have the remarkable difference.The highest pollen germination ratio in the morning 11:30,afternoon's germination ratio was low,was not suitable the pollination.â‘¢Coefficient of variation analysis,variance analysis and correlation coefficients analysis had been done on seven characters of twelve inbred lines such as plant height,plant crown,leaf length,leaf width,flower size,branch number and petal layer.The result indicated that the coefficient of variation of character were big at the low inbred line generation.As the generation added,coefficient of variation became smaller.The result of variance analysis indicated:the difference was not remarkable between different inbred lines,six characters except flower petal layer all have remarkable or the extremely remarkable difference,it was showed that the phenotypes of different inbred lines had a very big difference,the inbred lines had strong gene non-uniformity.Among seven characters of mose rose,only leaf length and leaf width had extremely remarkable correlation,leaf length and branch number had remarkable correlation,others characters had no correlation.Induction of polyploidyThis experiment used the colchicine solution to treat the different spots of mose rose plant organizations,mainly used three methods:induction of the germination seeds,the colchicine solution spot-out method,the improvement to soak the root to treat the mose rose separately,and the results indicated:â‘ The leaves of the doubling plants became longer and wider obviously,the flowers of the doubling plants became bigger and had more petal respectively.â‘¡Used the different colchicine concentrations to process mose rose germination seeds for 6 h,12 h,24 h and 48 h,finally discovered that processes under12h,0.05%concentrations,the biggest inductivity ratio was 11%;Used 0.05%, 0.10%,0.20%concentration of colchicine to treat mose rose stem growing point for 48h, when the colchicine concentrations was 0.10%,the inductivity was 75%,was the biggest, but used 4%DMSO and 0.05%colchicine mixed solution to induct mose rose root point, the inductivity ratio was only 16.7%.
