This thesis uses colchicine and trifluralin as inducers to conduct variants fromday Hemerocallis fulva callus tissue and adventitious bud through bath method and mixed cultivation method, and draw conclusions as follows:1. colchicine bath method works best on conditions of Hemerocallis fulva callus tissue's optimal concentration rate at 0.1% and 8 hours'treatment, with an inducement rate of 35.56%; Hemerocallis fulva adventitious bud's optimal concentration rate at 0.05% and 8 hours' treatment, with an inducement rate of 15.19%;2. mixed colchicine cultivation method works best on conditions of Hemerocallis fulva callus tissue's optimal concentration rate at 0.005% and 15days' treatment, with an inducement rate of 41.67%; Hemerocallis fulva adventitious bud's optimal concentration rate at 0.04% and 5days' treatment, with an inducement rate of 19.81%;3. trifluralin bath method works best on conditions of day Hemerocallis fulva callus tissue's optimal concentration rate at 0.01% and 12h hours'treatment, with an inducement rate of 22.22%; Hemerocallis fulva adventitious bud's optimal concentration rate at 0.01% and 6 hours' treatment, with an inducement rate of 21.75%;4. mixed trifluralin cultivation method works best on conditions of Hemerocallis fulva callus tissue's optimal concentration rate at 0.007% and 5days' treatment, with an inducement rate of 35.74%; day lily's adventitious bud's optimal concentration rate at 0.007% and 7 days' treatment, with an inducement rate of 13%;5. Set up an optimized system of Hemerocallis fulvachromosome sectioning technology. Using young root tip as testing material, treating it with dichlorobenzene preconditioning liquids of 4.5 hours, fixing with Carnoy's fluid for 24 hours at 4?, dissociating 10 mins in water bath with 1 mol/L hydrochloric acid and dying 20 mins with Carbol Fuchsin to get optimal chromosome tablet.6. According to the root tip chromosome tableting,all of the variantplants were aneuploids which have 25?29?31?39?40?50 chromosomes.7. Research on biological characteristics of induced variant Hemerocallis fulva. Variant Hemerocallis fulva has greener leaves with evident enlargement of lower epidermis stoma. Under same magnification, variant Hemerocallis fulva stomas' number only account for 42.44% of diploid ones', while leaf-shape-index of diploid Hemerocallis fulva is 2.21 times of tetraploid Hemerocallis fulva.
