Wolfiporia cocos(Schw.)Ryv.& Gilbn.,a higher basidiomycete,is a perennial saprophytic fungi,which belongs to Hymenomycetes,Aphyllophorales,Polyporaceae, Wolfiporia.its underground growth sclerotium is a traditional Chinese medicine nice nutritional value and medicinal value.The Wolfiporia cocos wild resources are very limited to the needs of the market.Although the artificial cultivation has been achieved, in Guizhou has not yet formed large-scale cultivation base,the degradation of strain is a bottleneck restricting the development of one of a series of studies on this subject,So The subject established initially the Wolfiporia cocos strains of rejuvenation system by the series of experiment,for the implementation to lay the foundation on the realization of Fuling Base-building and for large-scale production of Wolfiporia cocos.The experiment was studied,on mycelium morphology and nuclear fluorecent dyeing of the Wolfiporia cocos strains in microscopic observation.and the tissue of sclerotium were separated.The rejuvenation methods had used by ultraviolet light culture, tip mycelium culture,separating sclerotium and single spore isolation culture,The results showed the four Rejuvenation method can reach a certain rejuvenation effects,the combining of ultraviolet light culture and tip mycelium culture were the ideal method and means of rejuvenation.The optimum culture condition for inducing fruit bodies of wolfiporia cocos was studied under laboratory conditions from medium components,light intensity,temperature,culture time and generations.The optimum potato medium of agarose(PDA),temperature,light intensity,culture time,air relative humidity and generations for inducing fruit bodies of wolfiporia cocos are potato glucose agar,(27±1)℃.60%(the light Incubators about 12000LX).18~30d.about75%and 5~7 generations.respectively and the morphology of induced fruit bodies is including honeycomb.mountain.lab yrinth.small chrysanthemum.and needle shapes with the single or group.Monospore were isolated by diluting.Under 26 condition,spores at the highest rate 45%.Mycelia sprouted from single spores were denser,appeared white floss-shaped,had no evident mycelia in the air,the growth was slow.Mycelia sprouted from single spores had obvious different shape patterns,mycelia growth rate and the degree of dense of mycelia were obvious different.In the single spore mating test,all mating tests of spores from the same strain generated antagonism lines,among them,the minority of mating testes fromed fiabelliform region in the junction and the antagonism line disappeared in a short time.All mating testes of spores between different strains generated barrage.The same strain mating testes grew disharmoniously with each other.On the basis of the result,it is supposed that the two nuclei in the spores are heterogeneous and complementary,a single spore could germinate and develop into an individual.In the cultivation experiment,the quality of two times digging sclerotium was different to different strains.And the scend time digging sclerotium was more than the first time,and the quality of sclerotium was different each other,most strains' content of polysaccharide were in the range of 4.0%to 6.0%by Soxhlet extraction method.The content of polysaccharide was the highest to GZAAS1.0612 strain,about 8.08%.the next was 7.68%to GZAAS1.0611,GZAAS1.0603 is the lowest,about3.32%,the quality of sclerotium in research of Wolfiporia cocos strain cross breeding were between 4.42%and 5.91%.
