Preliminary Study On The Effects Of "Bait" On Yield And Quality Of The New Sclerotium In Wolfiporia Cocos

Wolfiporia cocos is commonly used in traditional Chinese herbal medicine. Its dried sclerotium is widely used in decoction of herbal medicine, Chinese patent drug and crude material of health care products etc based on its enormously officinal value. In this study,43strains of W. cocos were conducted for cultivation and the strain T1was used as bait sclerotium. The influence of cultivation used the fresh sclerotium as the bait on yield and quality of the new sclerotium in Wolfiporia cocos was researched systematacially. What is more, RAPD analysis of43Poria strains was developed to discuss the effect of cultivation by the genetic distance between the bait sclerotium and the others. The results were followed:(1) The yield, hardness, water content, polysaccharide content and triterpenoid content of poria cocos sclerotium were measured to investigate the influence of cultivation used the fresh sclerotium as a bait. As43strains of W.cocos were conducted for cultivation used the fresh sclerotium as a bait, there are43,9,26,32,18have an increase on yield、water content、hardness、polysaccharide content and triterpenoid content, respectively, compared with the control group without a bait.In these strains, there are42,1,21,27,12strains exhibited significant or very significant, respectively. And1.12kg averagely per cavern on yield, water content increased7%averagely per cavern; hardness increased11771g averagely per cavern, polysaccharide content increased2.03mg/g averagely per cavern and triterpenoid content increased0.87mg/g averagely per cavern. Otherwise, there are1,8,5,5,6strains exhibited not significant difference. However, there are0,34,17,11,25have an decrease on yield, water content, hardness, polysaccharide content and triterpenoid content, respectively, compared with the control group without a bait. In these strains, there are0,22,13,6,20strains exhibited significant or very significant, respectively. And water content decreased10.5%averagely per cavern; hardness decreased7989g averagely per cavern, polysaccharide content decreased1.71mg/g averagely per cavern and triterpenoid content decreased2.12mg/g averagely per cavern.(2) The RAPD analysis experiment was adopted to investigate the influence of the new sclerotium by the genetic distance between the bait strain and the others. When the value of genetic similarity coefficient is0.82,43strains could classify into4categories, with17,10,5,17strains and genetic similarity coefficient was0.82-1,0.79-0.87,0.73-0.79,0.70-0.81with T1, respectively. However, the results of the correlation analysis on genetic similarity coefficient showed that there was no significantly relevance between genetic similarity coefficient and the increase of average yield hardness, polysaccharide content, and the decrease of water content. There was a very significant correlation between triterpenoid content. T1, ACCC50478, ZJ, chuanjie NO.1, T1got the greatest increase on yield, water content, hardness, polysaccharide content and triterpenoid content, and the value of genetic similarity coefficient was1,0.791040,0.835821,0.835821,1with T1, respectively, compared with the control group without a bait. W.cocos NO.21, Z (1),901, Y1, NO.6got the greatest decrease on yield, water content, hardness, polysaccharide content and triterpenoid content, and the value of genetic similarity coefficient was0.7611940,0.940299,0.716418,0.880597,0.835821, respectively, compared with the control group without bait.In conclusion,43strains of W.cocos were conducted for cultivation used the fresh sclerotium as a bait,97.67%strains increased on yield significantly,79.07%strains decreased on water content,60.47%strains increased on hardness,74.42%strains increased on polysaccharide content and41.86%trains increased on triterpenoid content compared with the control group without a bait. And1.62kg averagely per cavern on yield, water content increased8.3averagely per cavern; hardness increased9828g averagely per cavern, polysaccharide content increased1.68mg/g averagely per cavern and triterpenoid content increased0.87mg/g averagely per cavern. The results of the correlation analysis on genetic similarity coefficient showed that there was no significantly relevance between genetic similarity coefficient of T1and the increase of average yield, hardness, and the decrease of water content. There was a very significant correlation on decrease of triterpenoid content. There was a significant correlation on increase of polysaccharide content.
