Ziziphus jujuba originating in China, has a long history of cultivation.It has been documented in 3000 years old so far. Jujube tree is one of the earliest cultivated in Henan province.More than two thousand years of varieties have Z.'huizao', Z.'bianhesuan', Z.'guangyangzao'and so on. Its has produced abundant hereditary variation in long-term culture.How use of these valuable germplasm resources more effective, will has a crucial role for the development of our province. By using ISSR markers technology supplemented by a survey morphologic index,to research genetic diversity, genetic structure and genetic relationship between Henan main cultivated Ziziphus jujuba varieties and Z.'huizao'Popolation.It provides a theoretical basis for breeding date and germplasm resources protection in our province.Main results were as follows:1. The establishment of the improved CTAB, is the ideal method of extracting DNA from the Zizi phus jujuba mature leaves.The major steps were removed the impurity before broken cells, splited to release DNA , precipitated and separated DNA,that this method could be well applied to ISSR reaction and other means of testing requirements.2.Optimum system of ISSR amplification for Chinese jujube. In 15μl reaction solution:Template 30ng;Primer 0.8μmol/L; 2×Taq PCR MasterMix 7.5μl;other's ddH2O. The amplified procedure was:Step1: 94℃5min;Step2:94℃45s,59℃45s ,72℃75s,40 cycles;Step 3:72℃10min, 4℃preservation. This experiment selects more than 10 polymorphism primers used in h izao colony and other eight varieties.3.Total 10 primers, were used in ISSR amplification of 117 huizao form and 8 other varieties forms, 96 loci were gained among which 87 loci were polymorphic. Average loci number amplified per primer of loci were 96, proportion of polymorphic loci was 90.62%.4. Henan Main Cultivated Ziziphus jujuba Varieties and Z.'huizao'Popolation of the genetic diversity analysis showed that: proportion of polymorphic loci of Henan Main Cultivated Ziziphus jujuba Varieties colony was 85.42%,average genetic distance was 0.3951, the number of alleles, the effective number of alleles , genetic diversity and Shannon information index in other 8 varieties population was 1.8542,1.4525,0.2720 and 0.4152;The form variety of the huizao colony, the standard deviation and variation coefficient of the leaf length, leaf wide and leaf shape index of the hui zao colony are: 0.89,0.50,0.30 and 18.24,21.01,14.42;proportion of polymorphic loci of huizao colony was 79.17%,average genetic distance was 0.2866, the number of alleles, the effective number of alleles,genetic diversity and Shannon information index was 1.7917,1.2941,0.1843 and 0.2903;5. The total populations genetic diversity of Henan Main Cultivated Ziziphus jujuba Varieties and Z.'huizao'Popolation was 0.3124, the inside populations genetic diversity was 0.2281,genetic diversity and coefficient of gene differentiation between two populations was 0.0843 and 0.2698, which indicated that 26.98% variation partitioned between populations.Nm<1(Nm=0.6766) mean that gene flow between two populations was too small to prevent genetic differentiation caused by genetic drift; Ziziphus jujuba'huizao'total populations genetic diversity was 0.1874, the inside populations genetic diversity was 0.1763,genetic diversity and coefficient of gene differentiation between two populations was 0.0111 and 0.0593, which indicated that 5.93% variation partitioned between populations. Nm>1(Nm=3.9662) mean that gene flow between two populations could prevent genetic differentiation caused by genetic drift.6. Genetic identity and genetic distance between populations was 0.7829 and 0.2448, Cluster analysis by Coefficient of genetic similarity to the nine varieties:huizao,lingbaodazao,guangyangzao,bianhesuan,tongbaidazao and changhongzao together first,and then get with the dongzao,wuhezao and jixinzao together.
