Adopting new kind of establishment and advanced micro-environment control technique for tissue culture are indispensable approach for automated industrial-scale tissue culture. But there are a lot of complexed problems about the environmental factors which efected the growth of plantlets,so it is urgent to develop environmental control system which adapt to use in plant tissue culture. To research the efect of micro-environment on culture plantlets,In this paper,The PHOTINIA×FRASERI plantlets were used as plant materials for experiments and were cultured in novel enveloped-shaped vessels,the Culture Pacd (CP),under light-emitting diode(LED)radiation resource, and the equipment of light and CO2 enrichment ect.for study on the effets of micro-environment such as CO2 concentration, light,sugar concentration on the growth of PHOTINIA×FRASERI plantlets.The results showed:(1)Using CP as cultivation vessel with the application of CO2 to compare the effect on the growth of Photinia Xfraseri Plantlets and its stomatal characters produced by various of sugar concentration(0g/L-1,10g/L-1,20g/L-1,30g/L-1 ).The results indicated that In point of a light intensity of 2000LX, a CO2 concentration of 3000ppm, the best sugar concentration for the cultivation of Photinia Xfraseri Plantlets was 0g/L-1 and 10g/L-1 ,it is obviously that in the case of CO2 concentration of a 2000ppm the Photinia Xfraseri Plantlets can grow normally by a photoautrophic mode , and it can increase the growth of the plantlet by improving the sugar concentration at some extent .(2)This experiment was done at a light concentration of 2000lx, without adding sugar to research the effect produced by different CO2 concentration (1000ppm,2000ppm,3000ppm,CK), the results indicated that : applying CO2 can obviously improve the growth status of Photinia Xfraseri Plantlets, the growth status of Photinia Xfraseri Plantlets at a CO2 concentration of 2000ppm was better than that of a CO2 concentration of 1000ppm, 3000 ppm and ck treatment , the effect of the treatment was worst as the CO2 concentration was 3000ppm, it showed that in this situation the better CO2 concentration was 2000ppm, too high concentration of CO2 prohibited the growth of Photinia Xfraseri Plantlets(3)This experiment compared the effect on the Photinia Xfraseri Plantlets produced by different light intensity (1000LX,2000LX,3000LX) with the application of CO2 ,the results showed that : improving the concentration of CO2 in the CP vessel and increasing the light intensity can obviously increase the growth of Photinia Xfraseri Plantlets, but the dry matter content,chlorophyll index,stomata ,and so on important determination indexes of the treatment at a light intensity of 1000LX was higher than that of a light intensity of 2000LX,3000LX.(4)This experiment used new type of LED as artificial light source to cultivate Photinia Xfraseri plantlets, the results indicates that : the culture plantlet under LED light has stronger photoauto-trophic ability compared with general daylight lamp the culture plantlet under LED light has stronger photoauto-trophic ability compared with general daylight lamp.The LED light at a ratio of 70%R+30%B can best increase the growth of the Photinia Xfraseri Plantlets compared to the general daylight lamp.
