Gushi Chickens were used as experimental materials.Different groups were vaccinated with Shigellae inactive vaccine cooperated by different amounts of interleukin-2(IL-2).Antibody titers in serum and indexes of immune organs were measured before and after immunization.Expressions of SIgA were studied and observed with immunohistochemistry technology .The results were analyzed through Qwin image analysis system.The following are the experimental results:⑴After immunization, antibody titers in serum are higher in the group vaccinated with Shigellae inactive vaccine cooperated by IL-2 than in the control group and the group vaccinated with Shigellae inactive vaccine.The difference is significant in the 2ed week.⑵After immunization, indexes of immune organs are larger in the groups vaccinated with Shigellae inactive vaccine cooperated by IL-2 than in the control group and the group vaccinated with Shigellae inactive vaccine.The difference is significant sometimes.⑶SIgA mainly appears in the secrecting cell membrane in the lamina propria.Some serecting cells lies in villi and some are around glands. Some SIgA appears in lamellae or is like-thread in glands.SIgA is secreted more with the time passing by.⑷SIgA is secreted around glands in the duodenum and in the lamina propria of mucosa and ville.SIgA increases with time passing by.Generally SIgA is more in the groups vaccinated with Shigellae inactive vaccine cooperated by IL-2 than in control group and the group vaccinated with Shigellae inactive vaccine .SIgA is secreted most in the Shigellae inactive vaccine+IL-2(50μg)group. SIgA locates around glands in the lamina propria of mucosa. in the interstuium jejumum. SIgA increases faster and is more in the Shigella+IL-2(50μg) group than in any other group.Most SIgA lies in the proper lamina of villi in grains in the interstuium ileum.IL-2 makes cells secret significantly more SIgA in the groups vaccinated with Shigellae inactive vaccine cooperated by IL-2 than in the control group and the group vaccinated with Shigellae inactive vaccine.SIgA mainly lies in clusters in the lamina propria of villi and around glands in the tonsillacecalis. Shigella+IL-2 can stimulate lymphocyte to secret more SIgA.SIgA mainly distributes in the lamina propria near villi base In the diverticulum vitellium.SIgA is expressed most in the Shigella+IL-2(50μg) group.In the interstinum rectum more SIgA appears in glands and most SIgA is in the Shigellae inactive vaccine+IL-2(50μg) group.⑸SIgA increases gradually in the gut as the time passes by and peaks in the 4th week.From then on it decreases slowly. ⑹SIgA is expresed most in the tonsillacecalis in the groups injected with IL-2 in the 1st week after immunization .However, SIgA is expresed most in the diverticulum vitellium in the control group and the group vaccinated with Shigellae inactive vaccine.Most SIgA appears in different organs in different groups in the 2nd week after immunization.There is most SIgA in the tonsillacecalis in the 3rd-7th week.⑺Shigellae inactive vaccine and IL-2(50μg) can stimulate lymphocyte to secret most SIgA.The results suggests that recombinant chicken interleukin-2 would be a potential adjuvant,immune enhancer and antibasterial agent.
