Hickory,with rich nutriments,can be refined oil and made dried fruit,but it is so waste that the large numbers of peels and dregs are discarded or may be used as fodders.The article studies about extraction and refining methods of Hickory leaf flavonoids and investigates the effect of its oxidation resistance.1.The extracting solution showed fuchsia through the qualitative test of hydrochloric acid-magnesium reduction reaction.The complex reaction of metallic salt reagent obviously produced color precipitate.The test of TLC was found that the extractive of Hickory leaf had the same yellow spotting with mtin in the same Rf.It demonstrated clearly that there had flavonoids in alcohol extraction of Hickory leaf.2.The ethanol was used in experiment to extract flavonoids from Hickory leaf,and yield was increased along with the increase of extraction temperature,extraction timing, ethanol concentration.By orthogonal test that Hickory leaf flavonoids in the highest rate of a group of technology for A2B3C1D2,corresponding to a rate of 18.9568%,the lowest rate in a group of combination is A1B1C1D1,a rate of only 8.1796%;At the same time, Hickory leaf flavonoids combination of the highest purity for A2B1C2D3,its purity is 13.0533%,the lowest purity is A1B1C1D1,only 8.8584%.Mudi Wu purity was.tested object and the optimum condition were:the concentration of 80%ethanol,feed than 1:20, the temperature of 90℃,from time to 6 h.3.Flavonoids purity was tested object,through several separation and purification methods,namely,lime milk-sulfuric acid,extraction,diatomite,macroporous resin,it was found that flavonoids purity by the macroporous resin was highest purity and the number was 59.63%.The lowest purity was obtained by diatomite,and the number was only 25.37%.For taking total flavonoids yield as tested object,the highest rates was 1.113% obtain by extraction method while the lowest rate was 0.638%obtained by lime Milk-Separation of sulfuric acid method.4.Through the test,it was found that the total flavonoids Hickory has the obvious antioxidant.(1) Determination of antioxidant activity tolard.It was shown that with the increase of total flavonoids,the upward trend of POV and acid value were slow down.It was shown from POV that 0.1%of the total flavonoids Hickory Group POV value was only 4.03(med /kg) after placed for 12days while that of blank Group exceeded 8.0(med/kg).The acid value was shown that adding 0.2%of the total flavonoids Hickory group of lard,acid value was only 4.8(kgKOH/g),while that of blank group of acid Rose was 8.2(kgKOH/ g).The results showed that Hickory leaf flavonoids possessed anti-lard rancidity role,and the ability was increasd with the increase antioxidant activity.(2) Anti-aging effects on mice.Aider Garage 42 days,it was concluded that Hickory leaf flavonoids could inhibit and slowed down aging of mice caused by D-galactose body, besides that it could increase SOD activity in serum,reduce MDA and decline MAO activity in.brain tissue.The high-dose gcoup produced highest SOD activity and the number was 167.10(U/mL),while the lowest aging model group,only 136.01(U/mL), Hickory leaf flavonoids could enhance the serum of mice SOD activity;by compared MDA in all groups,MDA of the highest aging model group was 18.95(nmol/mL), Hickory leaf flavonoids high-dose group was lowest and the number was 14.88(nmol/mL), Hickory description of the total flavonoids in mice with lower serum MDA in the role of the small group.The aging model small group MAO rat brain tissue in the highest, reaching 26.42(U/mg),Hickory leaf flavonoids high-dose group lowest,with only 13.63 (U/mg),Hickory leaf flavonoids that can inhibit MAO in the brain tissue of mice Activity.(3) The removal of flee radicals.1.0 mg/mL Hickory leaf flavonoids produced highest scavenging ability to hydroxyl radical(·OH) and the number was 63%,flavonoids concentration of 0.8 mg/mL group generated lowerest scavenging rate,flavonoids Concentration of 0.2 mg/mL group of hydroxyl radical(·OH) possessed lowest scavenging bility and its scavenging rate was only 22%.It was concluded that scavenging ability was increased with the concentration of total flavonoids.Content of 1.0 mg/mL flavonoids possessed highest scavenging bility to the superoxide radical(O2·-) and scavenging rate was 58%,when flavonoids was 0.2 mg/mL,the scavenging ability was lowest and the scavenging rate was only 11%.It was concluded from the results that total flavonoids could restrain(O2·-) and the scavenging ability was increased with the increase of flavonoids concentration.
