Existing alfalfa flavonoids extraction method and the leaf protein extraction phase asunder, usually in alfalfa hay as raw materials by using organic solvent to extract the flavones, because hay bulky, flavonoid content relative to the sophora japonica and other raw materials is low, the process on the industrialized feasibility is extremely low. Actually, alfalfa flavone mainly flavone glycosides, similar to that of alfalfa leaf protein leaching conditions, and the two, is different from the conditions of precipitation in the grass juice for alfalfa leaf protein associated with flavonoids extraction provides the theoretical feasibility. This topic focuses on the feasibility of industrialization, and explores the alfalfa leaf protein extraction technology of insert alfalfa flavonoids extraction program, without changing leaf protein extraction of raw materials (alfalfa grass), little changed leaching process on the, basis of appropriate leaf protein precipitation process, increase alfalfa flavonoids collection process, thus for alfalfa leaf protein extract and flavonoids. The process parameters leaching amount of flavonoids as 1.2245mg/g, the leaf protein extraction for 36.0956mg/g. Beating static method:material to water ratio, beating time, standing time three factor and four level orthogonal test, results of material to water ratio 17:510, beating time was 2min, the static set time 5h treatment combination is the best. The process parameters under leaching amount of flavonoids for 1.0955 mg/g, leaf protein extraction for 37.2451 mg/g.2. The coagulation method and technological parameters of leaf proteinCondensation heat:heat coagulation temperature, coagulation time of the two factor and four level orthogonal test, pulping and alkali leaching provided optimal combination by thermal condensed processing results to coagulation temperature of 50?, coagulation time of 5 min treatment is best, flavonoids retained amount 2.0088mg/g, leaf protein retained amount 32.9042mg/g. Beating static optimal combination by heat coagulation treatment, treatment results to coagulation temperature is 95?, the coagulation time of 15min treatment is best, flavonoids retained amount 0.4845mg/g, leaf protein retained amount 22.6708mg/g.Lime water condenses:lime addition amount of water, coagulation time and limewater concentration of three factors and four level orthogonal test, pulping and alkali leaching provided optimal combination with lime water coagulation treatment, the results with lime water concentration 0.825mg/mL, lime water adding quantity 6?L/10mL. Coagulation time 5min treatment combination for the best, flavonoids retained amount 2.0413mg/g, leaf protein retention amount 22.6662mg/g; beating static set optimal combination with lime water coagulation treatment. Results with lime water concentration 1.2375mg/mL, lime water adding quantity 10pL/10mL, condensed time 20min treatment combination for the best, flavonoids retained amount 3.6915mg/g, leaf protein retained amount 22.0599mg/g.3. The Collection method and technological parameters of flavonoids in brown liquidAcid precipitation pH, acid precipitation, temperature, acid precipitation time three factor and four level orthogonal test, beating alkali leaching optimal combination by thermal condensation optimal treatment and acid precipitation, results in acid precipitation pH and acid precipitation temperature of 30?, and acid precipitation time was 70min treatment combination for the best, flavonoids residues 0.3752mg/g, Ye Dan residual quantity 28.4542mg/g. Beating static optimal combination by lime water, condensed the best treatment and acid precipitation, results in acid precipitation pH and acid precipitation temperature of 30?, and acid precipitation time 45min treatment combination for the best, flavonoids residues 0.4328mg/g, leaf protein residues 4.6842mg/g.The test obtained fresh alfalfa combined extracting leaf protein and flavonoids in leaching, condensation, acid precipitation process of optimum process parameters, namely: material and water ratio of 17:510, beating time 0.5min, alkali provided pH9.8, alkali extraction time was 8min, lime water concentration 1.2375mg/mL, lime water adding quantity lO?L/ml, coagulation time 20min, acid precipitation pH and acid precipitation temperature of 30?, and acid precipitation time was 70min. In the series parameters leaf protein extraction rate for 14.036mg/g, flavonoids acquisition amount for 0.8493mg/g.
