Follicle-stimulating hormone(FSH) is a pituitary glycoprotein gonadotropin that is composed of two subunits,the commonα-subunit and the specificβ-subunit,which determines the binding to specific receptors and induction of biological response,and it plays a key role in hypothalamic-pituitary-gonad axis.FSHβis one of the most important hormone mainly related to reproduction in avian species.Many scientific studies indicated that FSHβgene was selected as the candidate gene for egg production.In this study,the Wanxi-White goose(n=112),a Chinese excellent mid-bodied spiece,and Rhine goose(n=64) were used to detect SNPs in the FSHβgene by PCR-SSCP.By genome comparison,2 pairs of primer was designed using Primer Premier5.0 and Dnastar programme to clone the partial sequence of Exon 3 and partial sequence of goose FSHβgene.PCR-SSCP method along with fragment sequencing was used in testing gene mutations at Exon3 site.Associations between mutation in FSHβgene and egg production were analyzed.Meanwhile,sequencing of partial Exon3 region and mutation detection at gene coding was also analyzed on Rhine goose to make comparison with Wanxi-White goose.The partial sequence of Wanxi-White goose and Rhine goose were cloned(GenBank accession number:EU252532).Homological analysis suggested that the homology of Wanxi-White goose partial sequence with Rhine and chicken partial sequence were 99%and64%,Wanxi-White goose exon 3 region with chicken exon 3 region were 95%,respectively.A mutation locus C-126T should be analyized.Genetic analysis suggested that,in the Wanxi-White goose populationm,the allelic frequencies detected by P2 primer were 0.87 and 0.13,respectively,three genotype(AA,AB and BB) frequencies were 0.78,0.19 and 0.03,respectively.x~2 fitness test suggested that the population was in Hardy-Weinberg equilibrium at the site.In the Rhine goose population,allelic frequecies detected by P2 were 0.26 and 0.74,respectively,genotype(CC,CD and DD) frequencies were 0.11,0.3 and 0.59, respectively.x~2 fitness test suggested that the population was in Hardy-Weinberg equilibrium at the site.In the Wanxi-White goose population,least square analysis showed that three genotypes detected by P2 primer had significant relation with egg production,the egg production for BB was higher (P<0.05) than that for AA and AB.In Rhine geese population,least square analysis showed that the egg production of CD and DD genotypes were higher(P<0.05) than that of CC genotype.P2 locus might be a molecular marker for reproduction trait.
