Understanding the genetic control of growth and fat deposition in goose will provide an opportunity for genetic enhancement of production performance and meat quality. Biological effects of growth hormone are abroad, including effects on cell differentiation, cell growth and the metabolism of protein, glucose and fat. The cytosolic fatty acid binding protein are involved in the intracellular fatty acid movement and transfer fatty acid from cell membrane to the site of fatty acid oxidation and glyceride as well as lecithoid synthesis. It plays an important role in fatty acid metabolism. The major objectives of the present study were to clone the GH and A-FABP genes, investigate the expression of the two genes, identify polymorphisms of the genes, and evaluate association between GH and A-FABP polymorphisms and traits of growth and body composition. Goose GH and A-FABP genes were cloned by RT-PCR method in the current research. RT-PCR and Northern Blot methods were developed to investigate the expression of the goose GH gene and A-FABP gene in heart, liver, lung, stomach, spleen, kidney, muscle, brain, fat, pituitary and testicle. The SNPs of goose GH gene were detected by the methods of PCR-SSCP and Direct sequencing. The correlation analysis between the polymorphisms of GH gene and growth and body composition traits in Rhine goose and Heilongjiang Zi goose were carried out using the appropriate model. These major results are as followed:(1) The cds, 3'UTR and intron2 of GH gene were cloned. The length of the cds of goose GH gene was 651bp, and the sequences has been submitted to the GenBank (Accession No: AF149895 and AF490903).(2) Two SNPs, C441A and T449C (Accession No: AF490903), were found in intron2 region of goose GH gene.(3) Population genetic analysis of two SNPs in the GH gene intron2 in Rhine goose and Heilonjiang Zi goose showed that the frequency of allele genotypes and gene between Rhine goose and Heilongjiang Zi goose were significantly different (p<0.05) .(4) Results of association analysis between the GH gene intron2 SNPs and growth and body composition traits in the various population showed that the body weight of six genotypes individuals derived from the two SNPs had significantly different in Rhine goose (p<0.05). The BB birds had significantly higher body weight than others, and CC birds had significantly lower body weight than others. In Heilongjiang Zi goose, the heart weight offour genotypes individuals derived from the two SNPs had significantly different (p<0.05). The AC birds had significantly higher heart weight than others, and BC birds had significantly lower body weight than others.(5) The result of RT-PCR and Northern methods showed goose GH gene was expressed only in pituitary and brain.(6) The cds and intron2 of A-FABP gene were cloned. The length of cds of the goose A-FABP was 399bp, and the sequence has been submitted to the GenBank (Accession No: A F472610 and AF442493).(7)RT-PCR results indicated that goose A-FABP gene highly expressed in fat tissue, heart, kidney, stomach, slightly expressed in liver, lung, brain, testicle, muscle, pituitary and didn't express in intestine and spleen.Candidate: Ao Jinxia Specialty: Animal Genetics & BreedingSupervisor: Prof. Li Hui...
