Study On The Technique System Of Micropropagation In Bletilla Striata

Bletilla striata is a perennial flowering plants in the Orchidaceae family.it is not only a medicinal herb,but also a precious flower.So,it already has been a high valuable ornamental,and has had a bright foreground on flower.Offshoot reproducing is the conventional breeding ways,which are inbenefical to commercialized breeding because of the low propagation coefficient.With the development of the technology of plant tissue culture,the tube seeding of Bletilla striata is obtained,There are many people who studying on propagatating Bletilla striata with tissue culture,and obtained great achievement,the micpropagation procedure of Bletilla striata had been researched,including the establishment of asepsis seeding,initial culture,subculture breeding,stronger nurslings breeding,rooting breeding,transplantion breeding.Addition to that,isolating and identifing of Bletilla striata endophytical fungi and fungi-concomitancy transplantion breeding had also been researchred.what had been researched are advantageous to the production of factory of Bletilla striata,and settling the basis of seedling technique of rhododendron.The seed of the Bletilla striata were as explants to establish the technich system of tissue culture.Multydesign and multyanalysis was used to fing out the best composition of medium and the best proceeding of culturing.The results were as follow:(1)bourgeoning conditions: temperature of the daytime is 20-25℃,temperature of the night is 18-20℃,culturing without light.Culture conditions:temperature 25℃,9-10 hours illumination everyday,and about 1500-2500Lux illumination intensity. (2)seed bourgeoning:The results indicated that the relationship between embryo age and the germination rate of the Bletilla striata was direct ratio.The germination rate of seeds with old embryo age was 100%,the germination rate of seeds with young embryo age was lower than older one.The medium can determine the germination rate of seeds with young embryo age,and the best medium for bourgeoning of the Bletilla striata seeds was MS,the germination rate can be 95%.(3)The best subculture medium was MS+6-BA 1.0mg/L+NAA 0.15mg/L.where the multiplication rate was 2.88.(4)The best rooting medium was 1/2 MS+ NAA 1.0 mg/L+banana 75g/L,Where the rooting rate could reach 100%,and the average of root of rooted plantlets was 5.5.(5) For transplantion of plantlets,the vermiculite:soil:perlite=1:2:1 was the best material,and the survial of plantlets could be up to 79% after transplanting soil.(6)A kind of endophytical fungi has been isolated from the root of the Bletilla striata.After rudiment identified,it was a Rhizoctonia genus fungi,and was thought to be Bletilla striata endophytical fungi.In fungi-concomitancy transplantion,the survial of plantlets could be up to 88% in the vermiculite:soil:perlite=1:2:1...
