| Objective This study was attempt to establish the regeneration system for B.striata through callus induction and determine rapid propagation methods for B.striata in vitro using liquid medium,then to research of B.striata polyploid induced with colchicines.Methods 1)Screening the optimal provenance of B.striata through consult literature material and scene inspection;2)The seed,stem,leaf and root tip of B.striata were used as explants to induction callus,then anti-browning and redifferentiation were researched.3)Using tissue culture seedlings of B.striata as material,to screen the optimum domestication conditions of B.striata.4)The seeds of B.striata were cultured in liquid medium,study the relationship of the optimum conditions and seed germination.5)The seeds of B.striata were sowed in different matrix after seeds germination,to screen the optimum direct seeding conditions of B.striata.6)To study on induced polyploid in B.striata with colchicines,then the diploid B.striata and the polyploid B.striata were be compared.7)Determinarion of polysaccharide in the B.striata by phenol-vitriolic colorimetry.Results 1)The optimal provenance of B.stiata was be found which was from Anhui Zhenyang Forestry Ecological Development Co.,Ltd.2)The optimal explant callus inducement of B.striata is seeds,MS+1.0 mg/L 6-BA+1.5 mg/L 2,4-D is the optimal medium for callus induction and MS+1.0 mg/L 6-BA+1.5 mg/L 2,4-D+15 mg/L ascorbic acid is the best medium for anti-browning.The optimal medium for redifferentiation is MS+0.1 mg/L NAA+0.5 mg/L 6-BA and the optimal medium for rooting is MS+0.8 mg/LNAA+1.0 mg/L 6-BA.Adding organics can promote the corm growth.3)The seedlings grow well in the nutrition soil,wich the root tip growth rate of seedlings is the highest.The nutrition soil is the optimal matrix for domestication,the yield of B.striata could be improved obviously by interplanting soybean and straw mulching.4)MS medium is the best medium for seeds germination.The best illumination time is 12h/d and the optimal pH is 5.8 to 6.5.5)The optimal matrix for direct seeding is humus soil mixed with vermiculite(1:1),spurting leaf-fertilizer affects the growth of B.striata significantly.6)We obtained the highest frequency of morphological variation when the seeds were treated with 0.1%colchicine for 7days.7)The plant treated with colchicine showed that have more polysaccharide than diploid one,the polyploid B.striata have the higher quality than diploid one.Conclusion Using seeds of B.striata as material,we established a callus induction and regeneration system and a direct seeding system of B.striata successfully,using these two system we can reproduction B.striata efficiently and quickly.And we get the higher quality B.striata by induced of polyploid with colchicines.This research has a big importance for utilization and conservation of B.striata. |
