| Objective:To analyze different expression proteins of the brucella melitensis vaccine strain M5 with a virulent strain 16M by the technologies of proteomics,and explore the mechanism molecular attenuation mechanism of the brucella melitensis vaccine strain M5 in whole level of proteins,and understand the mechanism of virulence in B.melitensis,and fred out the diagnosis markers of the brucellosis.Methods:Extracted total proteins of the brucella melitensis vaccine strain M5 and a virulent strain 16M by acetone cryochem,separated proteins by two-dimensional electrophoresis(2-DE),stained proteins by Coomassie brilliant blue,got the patterns, found out differential proteins,and obtained PMF(peptide mass fingerprinting) by MALDI-TOF-MS,gained related the information of proteins by MS-Fit database.Results:In this study,2-DE Patterns with high resolution and reproducibility from the proteins of the strains M5 and 16M was obtained.The total Protein distribution pattern of the strains M5 and 16M 2-DE is very similar.A total of 797±22 and 831±29 protein spots were detected for strains 16M and M5,respectively.And the matching rate of them is 71.25%.49 out of 53 Proteins Pots standing for 35 different ORFs were identified by the technologies of mass spectrogram with bioinformatics, of that 27 Proteins were up-regulated and 22 Proteins were down-regulated in the strain 16M.49 proteins pots identified by the technologies of mass spectrogram with bioinformatics seem to play important roles in a variety of pathways including sugar binding,protein production,lipid metabolism,cell skeleton,and molecular chaperone. Infer the DNA protection during starvation,Omp25,Sugar-binding protein maybe concerned with the mechanism of virulence upexpressed proteins in strain 16M.The Invasion Protein B maybe concerned with brucella pathogenicity expressed proteins both in strain M5 and 16M.The 31 kDa outer-membrane immunogenic protein precursor maybe concerned with brucella immunogenic upexpressed proteins in strain M5,so it is significant to explore DNA vaccine with Omp31. |
