Different transplanting and topping time was used in this research to study its effect on the chemical composition and tissue structure of E-yan 1 in Hunan province. The results were as follows:1. The single leaf weight decreased as the transplanting time postponed. While the topping time postponed, the single leaf weight also decreased.The single leaf weight of the middle leaf was heavest, and the lower leaf was least.2. The protein content of the upper leaves were influenced largely by the transplanting and topping time, while the middle and lower leaves were influenced slightly. As the leaf deposition rised, the protein content increased. Whereas the content of reducing sugar was decreased as the transplanting and topping time postponed.3. The content of nicotine decreased with the postpone of transplanting and topping time. And as the leaf deposition rised, the content of both nicotine and total nitrogen increased. As the topping time postponed, the content of total nitrogen exhibited reversed V curve. While the transplanting time postponed, except for the emerge-budding the content of total nitrogen exhibited V curve.4. Early transplanting of the same topping time and topping at flowering stage at the same transplanting time could increase the content of K~+. The content of Cl~- of upper and lower leaves didn't change by the different topping time; however, the middle leaves were changed as the topping time postponed.5. As the transplanting time postponed, the thickness of leaf, palisade tissues, spongy tissues of upper and lower leaves all turned thinner. Otherwise, the changing of thickness of leaf and spongy tissues of middle leaves were the same: The thickness of plants transplanted at April 26th was the largest, and which transplanted at April 16 was the lowest. Along with the topping time postponed, the thickness of leaf, palisade tissues, spongy tissues and the thickness and width of upper and lower epidermis cell all turned thinner, moreover, the lower leaf changed most, middle leaf least.6. The changing of the thickness of palisade tissues, the ratio of tissues, the ratio of the thickness of palisade tissues to leaf of upper leaves and the ratio of space of lower leaves were the same in which April 16th showed the largest phynotypic values, while April 6th showed the lowest values. The changing of the ratio of tissues, the thickness of palisade tissues to leaves of lower leaves and the ratio of space of upper leaves were all decreased as the transplanting postponed. With the topping time postpone the ratio of space exhibited V curve.
