Effects Of 'Yimu Shenghua San' On Content Of Cytokines In Placental Tissues Of Cows Affected With Retention Of Placenta

Abstract/Summary:
In this study, through the enzyme-linked immunosorbent assay (ELISA) detection of placental tissue of IL-2, TNF-α, IFN-γ, IL-4, IL-10 content changes, microscopic detection of afterbirth placenta of pathological changes, the clinical effects that'Yimu Shenghua San'preventing and treating the retention of fetal membrane in cows were observed, to study the the occurrence mechanism of the placenta retention and the prevention and treatment mechanism of the traditional Chinese herbal medicine'Yimu Shenghua San'. The cows were divided into 4 groups. Gestational times 2-4 randomly selected 10 cases of healthy Holstein, as a control group (group A); Select gestational times 2-4 post-natal health of Holstein 17 cases as prevention group (group B), received a gavage of'Yimu Shenghua San'250g after parturition immediately; group C as placenta retention, group D as treating placenta retention that couldn't discharge the fetal membrane after 12h of parturition was received an oral gavage of'Yimu Shenghua San'500g. The results showed that the'Yimu Shenghua San'had good effects on prevent the cows placenta retention, the effective rate was 92.59%, the incidence rate was 7.41%, the incidence of endometritis rate was 0; The medicine had a significant therapeutic effect in preventing placenta retention, the effective rate was 88.46%, the incidence of endometritis rate was 19.23%. The contents of IL-2 in placental tissue lysate of group A was similar to that of group B (p>0.05). The contents of IL-2 in placental tissue lysate of group D was significantly higher than those of group A and group B (P <0.01). The contents of IL-2 in placental tissue lysate of group C was significantly higher than those of group A, group B and group D (P <0.01). The contents of TNF-αin placental tissue lysate of group A was higher than those of group B (P <0.05). The contents of TNF-αin placental tissue lysate of group A was similar to that of group D (P> 0.05). The contents of TNF-αin placental tissue lysate of group B was similar to that of group D (P> 0.05). The contents of TNF-αin placental tissue lysate of group C was significantly higher than those of group A, group B and group D (P <0.01). The contents of IFN-γin placental tissue lysate of group A was higher than those of group B and group D (P <0.05). The contents of IFN-γin placental tissue lysate of group B was similar to that of group D (P> 0.05). The contents of IFN-γin placental tissue lysate of group C was significantly higher than those of group A, group B and group D (P <0.01). The contents of IL-4 in placental tissue lysate of group A was similar to that of group B (P> 0.05). The contents of IL-4 in placental tissue lysate of group D was significantly lower than that of group A and group B (P <0.01). The contents of IL-4 in placental tissue lysate of group C was significantly lower than those of group A, group B and group C (P <0.01). The contents of IL-10 in placental tissue lysate of group A, group B and group D were no significant difference (P> 0.05). The contents of IL-10 in placental tissue lysate of group C was significantly lower than that of group A, group B (P <0.01) and was similar to group D (P> 0.05). The results indicate that it has closely relattionship between the placenta retention with the contents of IL-2, TNF-α, IFN-γ, IL-4 and IL-10 in placental tissue lysate, and the pathological changes of placenta.'Yimu Shenghua San'have a good effect on preventing and treating the placeta retention,'Yimu Shenghua San'can regulate the secretion of placental cytokine.
