| Acer mono Maxim.is an excellent tree species for ornamental,medicinal and timber purpose.But its application was hindered for lack of detailed study of propagation biology.The water permeability of seed coat,bio-effects of seed extraction solution,variation patterns of seed storage substances and its related enzymes,and content and balance of endogenous hormones in seeds during low-temperature stratification were studied taking A.ginnala.The results were shown as follow.1.The seedcoat of A.mono and A.ginnala has an excellent character of water absorption. So it may be not the main cause of seed dormancy.Seed extraction solution of A.mono and A. ginnala has strong inhibition effect on seed germination of Chinese cabbage,the intensity of the inhibition effect was reduced along with the increase of stratification days.The inhibition intensity of A.mono seed extraction solution was stronger than that of A.ginnala during stratification and 80 days stratification is not enough for complete release of the inhibition effect both for A.mono and A.ginnala as well.2.Changing patterns of storage substances and its related enzymes in seeds of A.mono and A.ginnala during low-temperature stratification. During seeds stratification to break dormancy,the content of starch and soluble protein in the A. mono and A.ginnala seeds reduces before increased,but the content of fatty acid in the A. mono seed reduces gradually,the content of fatty acid in the A.ginnala seed reduces before increased,The total activity of the lipase and amylase volatile,this change was lingering to degradability of fatty acid amylum and soluble protein.3.The content and perpotion of GA and CTK was increased and that of ABA was decreased during low-temperature stratification.During seed stratification,the content of ABA in A.mono and A.ginnala seeds reduced gradually,and the contents of GA and CTK increased at the same time,IAA contents have a complex changing pattern.The ratios of GA/ABA, IAA/ABA and CTK/ABA in A.ginnala seeds increased ultimately,and that in A.mono seed changed smoothly.These results can explain to some extent the reason that the inhibition intensity of A.mono seed extraction solution was stronger than that of A.ginnala. |
