The updating and modify of ABC model of floral organ development show plant MADS-box gene plays the important regulate role in the process of floral development.Genic repeat, functional differentiation and network interaction result in the variety of angiosperm floral form,common analysis from single model plant to multi-plant and comparative study are contributed to elucidate the development of floral traits and the diversity of molecular mechanisms.Eustoma grandiflorum is the herbage ornamental plant,which pertains to gentian.There are cultivars with single and double flowers.In double flowers,the developments of four whorl organs are normal and the number of stamens is stable,which is different from petal-like stamen phenomenon in many double flowers.In order to elucidate the molecular mechanism of floral development,the following investeigation was made on C/D MADS-box gene:1.Cloning of MADS-box class C and D genes in Eustoma grandiflorumC-function gene EgPLEl and D-function genes EgMADSl were cloned by the method of 3'-RACE and the technology of RT-PCR,respectively.The total RNA at different developmental stages was used as templet,which primers are gene-specific primers.C-terminal of the two genes have conservative AG motifâ… and AG motifâ…¡which belong to AG-like subfamily.The results of Southern blot show EgPLE gene has multi-copy and EgMADSl gene has four copies in genome.2.Expression analysis of MADS-box class D genes in Eustoma grandiflorumStudying on the expression of different stages during Eustoma grandiflorum floral development using Northern blot,the result shows EgMADSl gene expression is detected only in carpels and ovule.3.Establishment of genetic transformation vector of EgMADS1 geneIn order to validate function of MADS-box class D genes in Eustoma grandiflorum,the EgMADSl recombinant expression vector and RNAi vector have been constructed using Gateway technology and was transferred into Agrobacterium LBA4404 strain in tobacco.
