With the foundition of the technology of kelp gametophyte clone, kelp seedlings clone has been applied widely. The seawater must be disinfected before using in the kelp gametophyte clone, and the method of boiling is widely used. But it will take lots of energy and labors to boil seawater in the culture process of the large-scale kelp gametophyte clone. Therefore, it is a very significance thing to obtian a disinfect seawater method which was more cheaper and convenient for the large-scale culture of the kelp gametophyte clone.The five methods of boiling, hydrochloric acid, sodium hypochlorite clone, ultraviolet radiation and ozone were chosen to disinfect the seawater. The DO, Alkalinity, chlorinity, salinity, pH,nutrients,CODMn,BOD5 and trace element of theose disinfect the seawater were determined; and the effects of different disinfection seawater on the growth of kelp gametophyte clone of Laminaria japonica were researched.The results showed that only the content of Fe and pH in five disinfectant seawater had difference. The content of Fe in boiling water was lower, however, which was higher in HCl disinfectant seawater; The pH of NaClO disinfectant seawater was the lowest in five disinfectant seawater, which was only 7.48. The growth of the kelp gametophyte clone was the fastest in boiling seawater, followed by the HCl disinfectant seawater, the NaClO disinfectant seawater was the worst one. A higher development rate of kelp gametophyte clone was obtianed in the boiling seawater and ultraviolet seawater. And the development rates of kelp gametophyte clone in the HCl disinfectant seawater and ozone were slightly lower than that of the above two disinfectant seawater. However, the development rate of the kelp gametophyte clone in the NaClO disinfectant seawater was extremely slow, even non-development was found. It was clearly that the disinfection of seawater with HCl could be used in the cultivation of kelp gametophyte clone. Diatoms were found in disinfectant seawater with ultraviolet and ozone, which indicated that the seawater not thoroughly disinfected, therefore the further researches need to be carried out for large-scale production.
