In this experiment,the immunohistochemistry was made use of to position the soybean allergic proteins in carp intestines and investigate the distributing of the soybean allergic proteins in carp intestines.The absorbing of allergic macromolecules was disclosed to some extent.At first,700 concinnous carps were chosen and bred in the fish bowl for 15 days.The stoyer in the experiment was randomly divided into 5 dealing groups.Each group was fed by DSBM instead of 0%,15%,30%,45%and 60%fish meal and set up 3 repetitions with the 8 weeks feeding cycles.8 weeks later,the carps were totally killed,tissue samples of proximal intestine,middle intestine,distal intestine were taken and paraffin section was made after fixed by paraformaldehyde.Glycinin andβ-conglycinin in each part of intestine tissue were positioned through immunohistochemistry method,and the changing of glycinin andβ-conglycinin in intestine tissue was investigated through making a comparison.The resuks of IOD of glycinin in intestinal tract showed that the IOD in distal intestine was much more than in Proximal intestine.But the difference of IOD between Proximal intestine and Middle intestine was not conspicuous.It was also showed that during the different groups, the IOD in Proximal intestine was not conspicuous.In middle intestine,the integral optical density of group Containing 60%DSBM was extremely higher than the group Containing 15% DSBM(P<0.01),30%DSBM and 45%DSBM group(P<0.05);In distal intestine,the integral optical density of group Containing 60%DSBM was extremely higher than the group Containing 15%DSBM(P<0.01),30%DSBM and 45%DSBM(P<0.05),while there is no significant difference in the group Containing 45%DSBM.The integral optical density of group Containing 45%DSBM was extremely higher than the group Containing 15%DSBM and 30%DSBM group(P<0.05).The results of integral optical density ofβ-conglyinin in carp intestines showed that the integral optical density in distal intestine was more than the one that in middle intestine,and the one in middle intestine was also more than that in proximal intestine.During the different groups,in proximal intestine,the integral optical density of 60%DSBM group was extremely higher than the group Containing 15%DSBM(P<0.01) and the group Containing 30%DSBM (P<0.05),while there was no significant difference between the 45%DSBM group and the 60%DSBM group.In middle intestine,the integral optical density of group Containing 60% DSBM was extremely higher than the group Containing 15%DSBM,30%DSBM(P<0.01) and the group Containing 45%DSBM(P<0.05);in distal intestine,the integral optical density of group Containing 60%DSBM was extremely higher than the one that of the 15%DSBM group(P<0.05),30%DSBM group and the 45%DSBM group(P<0.05).The mean of optical density of the glycinin in different intestinal tract had great difference(P<0.01).The mean of optical density in distal intestine was much more than the one in middle intestine;the mean of optical density in middle intestine was much more than the one in Proximal intestine.But among the different groups,the difference was not conspicuous.The average optical ofβ-conglycinin in Distal intestine was more than the one in middle intestine and proximal intestine.And the difference between Proximal intestine and Middle intestine was not conspicuous.Among different groups,the difference was not conspicuous.The integral optical density and the mean of optical density of glycinin andβ-conglycinin in the intestinal plica was much more than the ones that in crypt(P<0.01).In all,with the increasing of the DSBM's amount,the changes of the soybean allergic proteins amount in carp intestines was stable first and then increase,whileβ-conglycinin upgraded placidly.The amount of the group of the soybean allergic proteins andβ-conglycinin in distal intestine were more than the ones that in proximal intestine and middle intestine.With the increasing of DSBM,the highth of plica in carp-intestines was obvios decline.And the foregut,midgut and hindgut also had the tendency of decline.
