Totally 250 Chinese Holstein were used as research populations,PCR,enzyme cutting and sequencing analysis were carried on four polymorphic sites of lactoferrin gene by PCR-RFLP technology,three kinds of genotypes were discovered existing on these four polymorphic sites;the association between these sites' polymorphisms and the mastitis was analyzed with the breeding material record and the performance trait target using the general linear model(GLM),and the effect examination between different genotypes generated on-916 location and-927 location with production character was not remarkable,but the different genotype's performance trait target generated on+72 location and-478 location was different,the mastitis resistant genotype and the prevalent allele were found,and it provided the powerful basis for the further molecular assisted selection,and provided the theory basis for the cow mastitis's disease-resistant breeding work.results were as follows:Four polymorphic sites were definited by lactoferrin gene registered on NCBI and according to Seyfert's report,PCR and the sequencing analysis were carried on,and the polymorphism were discovered on these four sites,these were G to A divert at-927 location,T to G divert at-916 location,G insert at-478 location and T to C divert at+72 location,there was consistency between anticipated result with report.TagⅠ,HaeⅢ,EcoT14Ⅰand HpyF10ⅤⅠwere carried on PCR production of-927 location,-916 location,+72 location and-478 location,three genotypes on these four polymorphic sites were discovered by polyacrylamide gel electrophoresis detection and analysis.And there were genetype AA,genetype AB and genetype BB.By the performance trait data which the DHI determination records,the general linear model was used to significance test between different genotypes and SCS,milk yield,protein percentage,fat percentage,Result indicated:the genotype AB individual's SCS on+72 location(T/C)was extremely and remarkably lower than the genotype BB individual(P<0.01),obviously lower than the genotype AA individual(P<0.05),the difference between the genotype BB individual and genotype AB individual was not remarkable;At the same time,the genotype AB individual's milk production was obviously higher than the genotype AA,the genotype BB individual(P<0.05),indicated that the genotype AB individual appears mastitis resistance;the genotype AA individual's SCS on-478 location(-/G)was extremely and remarkably lower than the genotype AB and BB individuals(P<0.01),and the genotype AA individual's milk production was obviously higher than the genotype AB,the genotype BB individual,indicated that the genotype AA individual appears mastitis resistance;In addition,the genotype AB individual's protein percentage and fat percentage on-478 location(-/G)were obviously higher than the genotype AA and BB individuals.Therefore,the allele A was prevalent.The effect examination between different genotypes generated on-916 location(T/G)and-927 location(G/A)with SCS,milk yield,protein percentage,fat percentage were not remarkable.
